摘 要:目的探讨含红芪和含黄芪的益气养血汤抗免疫衰老的可能作用机制并比较两者的作用效果。方法 30只青龄昆明种小鼠随机分成红芪益气养血汤组、黄芪益气养血汤组、空白组,每组10只,红芪益气养血汤组及黄芪益气养血汤组分别给予相应生药6 g/(kg·d),灌胃量为0.4 ml/(20 g·d),空白组灌胃相同体积生理盐水,连续灌胃14天,末次灌胃2 h后眼球摘除取血,制备益气养血汤含药血清和空白血清。调整SAMP8小鼠脾脏T淋巴细胞浓度为所需浓度,分为红芪益气养血汤组(含药血清)、黄芪益气养血汤组(含药血清)、空白血清组(空白血清)、青龄鼠组(完全培养液)、SAMP8组(完全培养液)。培养72 h后,检测各组细胞增殖能力,T淋巴细胞表面分子包括CD8^+、CD4^+、CD4^+/CD8^+、CD28^+CD152^+表达情况,以及细胞培养上清液中白细胞介素2(IL-2)和γ干扰素(IFN-γ)水平和细胞中PI-3K m RNA表达、β-半乳糖苷酶染色细胞阳性率。结果与青龄鼠组比较,SAMP8组小鼠脾脏T淋巴细胞增殖能力、细胞表面CD8^+分子表达、培养上清液IL-2和IFN-γ水平、细胞中PI3K m RNA表达明显降低,CD4^+、CD4^+/CD8^+、CD28^+CD152^+双阳性表达及β-半乳糖苷酶衰老细胞阳性率均明显升高(P<0.05)。与空白血清组比较,红芪益气养血汤组和黄芪益气养血汤组均能提高T淋巴细胞的增殖能力、培养上清液中IL-2和INF-γ水平和细胞中PI3K m RNA表达,减少β-半乳糖苷酶染色细胞阳性率(P<0.05),并且在改善细胞增殖能力、提高IL-2水平及PI3K m RNA表达方面红芪益气养血汤组优于黄芪益气养血汤组(P<0.05)。结论含红芪和含黄芪的益气养血汤含药血清均能增强SAMP8小鼠脾脏T淋巴细胞的体外增殖能力,可能通过促进IL-2及IFN-γ分泌、上调PI3K m RNA表达量、降低β-半乳糖苷酶表达,从而改善T淋巴细胞的免疫衰老状态,并且含红芪的益气养血汤作用效果优于含黄芪的益气养Objective To discuss and explore possible mechanism of Yiqi Yangxue Decoction(益气养血汤) containing Hongqi( Radix Hedysari 红芪) or containing Huangqi( Radix Astragali,黄芪) against immunosenescence,and to compare the effect of both. Methods Randomly divided 30 Kunming mice of young age into the Hongqi Yiqi Yangxue Decoction group,the Huangqi Yiqi Yangxue Decoction group and the blank group,with 10 mice in each. The Hongqi Yiqi Yangxue Decoction group and the Huangqi Yiqi Yangxue Decoction group were both given 6 g /( kg·d) of corresponding crude drugs,with 0. 4 ml /( 20 g·d) of gastric volume. The blank group was given with equivalent saline in gavage,continuing for 14 days. Mouse eyeballs were removed to draw blood 2 hours after the last gavage to prepare serum containing Yiqi Yangxue Decoction and blank serum. Density of T-lymphocytes in spleen of SAMP8 mice were regulated to the density needed,and a Hongqi Yiqi Yangxue Decoction group( drug serum),a Huangqi Yiqi Yangxue Decoction group( drug serum),a blank group( blank serum),a young age mice group( absolute nutrient solution) and a SAMP8 group( absolute nutrient solution) were set up for T-lymphocytes culture.Proliferation ability of each cell,the expression of T-lymphocytes surface molecule including CD8~+,CD4~+,CD4~+/CD8~+and CD28~+CD152~+,levels of Interleukin 2( IL-2) and gamma interferon( IFN-γ) in cell culture supernatant,the expression of PI-3K m RNA and positive rate of β-galactose glucoside enzyme staining in cells were examined after cultivating for 72 hours. Results Compared with those in the young age group,cell proliferation ability,the expression of CD8~+molecule on the cellular surface,level of IL-2 and IFN-γin cell culture supernatant and the expression of PI3 K m RNA in cells of T-lymphocytes in spleen of mice remarkably decreased,while double positive expression of CD4~+,CD4~+/ CD8~+,CD28~+CD152~+and positive rate of β-galactose glucoside enzyme aging ce
