摘　　要：目的:探讨华蟾素对直肠癌SW480细胞的抑制作用及其机制。方法:以不同浓度的华蟾素作用于人直肠癌SW480细胞,用MTT法检测华蟾素对直肠癌SW480细胞增殖的影响;流式细胞术(FCM)观察华蟾素对细胞周期的影响;划痕实验检测华蟾素对直肠癌SW480细胞迁移能力的影响;蛋白印迹法(Western blot)检测华蟾素对凋亡蛋白Bcl-2表达的影响。结果:MTT法显示华蟾素显著抑制直肠癌SW480细胞增殖,其抑制率呈时间及浓度依赖性;FCM检测发现华蟾素使直肠癌SW480细胞的G2/M期延长,S期缩短,周期主要阻滞在G2/M期;划痕实验结果显示:华蟾素可降低直肠癌SW480细胞的迁移能力;Western blot结果显示华蟾素可使Bcl-2蛋白表达水平降低。结论:华蟾素能够抑制直肠癌SW480细胞的生长,并阻滞细胞周期在G2/M期,抑制其迁移,其机制可能与影响凋亡蛋白Bcl-2的表达相关。Objective： To study the effects of Cinobufotalin on the growth of human rectal cancer cell lines SW480,and the possible mechanism. Methods： The inhibitory effect of Cinobufotalin on cell proliferation was examined by MTT assay and cell cycle was detected by flow cytometry（ FCM）. Wound healing test was used to measure the migration ability of SW480 cells. The expression level of Bcl- 2 protein levels were detected by Western blot. Results： The Cinobufotalin singnificantly inhibited cell proliferation,blocked the cell cycle in the G2/ M phase. The migration capacity of Cinobufotalin- treated cells was decreased compared with that of the negative control cells. The expression levels of Bcl- 2 protein were down- regulated when SW480 cell were treated by Cinobufotalin. Conclusion： Cinobufotalin singnificantly inhibited cell proliferation,blocked the cell cycle in the G2/ M phase,and decreased the migration ability of SW480 cells signifycantly. These effects may be related with its function in down- regulating Bcl- 2 protein.
