摘  要:为研究灵芝酸对异烟肼所致免疫性肝损伤的保护作用,采用180 mg/kg异烟肼灌胃制备家兔免疫性肝损伤模型。罗氏全自动生化仪测定血清中丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)含量;HE染色法观测家兔肝脏病理学变化;ELISA法检测血液中干扰素(INF-γ)和肿瘤坏死因子(TNF-α)含量变化;分光光度法测定肝组织中丙二醛(MDA)的含量。结果为:异烟肼20 h可诱导家兔免疫性肝损伤血清中ALT和AST的含量分别为136 U/L和248 U/L;灵芝酸干预后ALT和AST分别为67 U/L和126 U/L;HE染色法观察灵芝酸干预后肝脏尚存少量的空泡;INF-γ和TNF-α分别从26.41 pg/m L和21.86 pg/m L下降为22.67 pg/m L和18.82 pg/m L;MDA从3.54μmol/L下降为1.5μmol/L。灵芝酸对免疫性肝损伤的保护作用可能是通过下调细胞因子分泌、降低肝脏MDA水平、抑制脂质过氧化反应来完成。Objective: to study the protective effect of Ganoderma lucidum on immunological liver injury by isoniazide. Methods: Immunological liver injury model of rabbits were induced by being given isoniazid 180 mg / kg by gavage. The ALT and the AST were assayed by Roche automatic biochemical analyzer. Histopathologic changes of the hepatic tissues were observed by the HE stained method in rabbit. The serum INF- γ and and TNF- α levels were determined by ELISA. The content of MDA in serum was measured by spectrophotometry. Results: The ALT level decreased from 136 U / L to 67 U / L and the AST level was from 248 U / L to 126 U / L by Ganoderma lucidum on 20 h. A small amount of vacuolar degeneration were observed on hepatic tissues after Ganoderma lucidum intervention. INF- γ level decreased from 26. 41 pg / m L to 22. 67 pg / m L,TNF- α level was from 21. 86 pg / m L to 18. 82 pg / m L. MDA form 3. 54 μmol / L to 1. 5 μmol / L. Conclusion: The protective effect of Ganoderma Lucidum on immunological liver can be probably completed by regulating cytokines secretion,reducing MDA level and by inhibiting lipid peroxidation reaction.
