摘 要:目的:观察壮骨止痛方对去卵巢骨质疏松大鼠血清TGF-β和AKP3的影响,探讨其抗骨质疏松机制。方法:72只SD大鼠按体质量随机抽出假手术组12只,造模组60只,造模组采用双侧去卵巢法造模,假手术组只切除卵巢周围相应质量的脂肪。造模成功后,大鼠按体质量随机分为模型组、壮骨止痛方高剂量组(13.2 g/kg)、壮骨止痛方中剂量组(6.6 g/kg)、壮骨止痛方低剂量组(3.3 g/kg)和戊酸雌二醇组,每组12只。术后第5天开始药物干预,模型组和假手术组给予相应体积的纯净水,持续13周。给药结束后,检测大鼠骨密度、血清TGF-β和AKP3含量。结果:模型组大鼠血清AKP3显著高于假手术组,骨密度低于假手术组,差异均有统计学意义(P<0.01)。壮骨止痛方高、中、低剂量组大鼠血清AKP3含量显著低于模型组,骨密度显著高于模型组,差异均有统计学意义(P<0.05或P<0.01)。结论:壮骨止痛方可调节去卵巢骨质疏松大鼠TGF-β和AKP3,可能是其抗骨质疏松的作用机制之一。Objective: To explore the effect of Zhuanggu Zhitong Prescription (壮骨止痛方, ZGZTP) on TGF-β and AKP3 in the serum of ovariectomized rats with osteoporosis, and to investigate its mechanism in anti-osteoporosis. Methods: The 72 rats were randomly divided into sham group (12) and model group (60), the model group rats were ovariectomized and the sham group rats were excised equal weight fat. After the modeling, the rats were randomly divided into model group, ZGZTP high-dose group, ZGZTP middle-dose group, ZGZTP low-dose group and estradiol valerate group, 12 rats in every group. 5 days after the operation started to drug therapy for 13 weeks. The model group and sham group rats were given equal volume water. After the test, detected the content of TGF-β and AKP3 in the serum. Results: Compared with the sham group, the content of AKP3 in the serum of model group was significantly increased (P〈0.01), and BMD was significantly declined (P〈0.01). There was an increasing trend in the content of TGF-β, but the difference had no statistically significant (P〉0.05). Compared with the model group, the content of AKP3 in the serum of ZGZTP groups was significantly declined (P〈0.01), and BMD was significantly increased (P〈0.01). There was an declining trend in the content of TGF-β, but the differenee had no statistically significant (P〉0.05). Conclusion: ZGZTP can adjust the secretion of TGF-β and AKP3, and this may be one meehanism of ZGZTP in anti-osteoporosis.
