摘  要:目的建立用于测定复方芍红颗粒中芍药苷含量的HPLC法。方法采用色谱柱Kromasil C18(4.6 mm×250 mm,5μm),以甲醇-水(40∶60)为流动相,流速1.0 m L/min,检测波长232 nm,柱温30℃。结果芍药苷检测浓度在0.008~0.512 mg/m L范围内与峰面积呈良好线性(r=0.9998);平均回收率为100.3%,RSD为1.38%;精密度、稳定性和重复性试验RSD均小于2%。含量测定平均值为17.54 mg/g。结论该方法简单、快速,精密度高,重复性好,所建立的方法可以用于复方芍红颗粒中芍药苷的质量控制。Objective To establish an RP-HPLC method for the determination of the content of paeoniflorin in the Fufangshaohong Granules. Methods The chromatographic columns(Kromasil C18, 4.6 mm×250 mm, 5 μm) was applied to test the content of paeoniflorin. A mixture of methanol-water(40∶60) was utilized as the mobile phase whose flow rate was1.0 m L/min. The detection wavelength was 232 nm, and the column temperature was set at 30℃. Results The results showed there were linear relationships between peak area and concentration of paeoniflorin within 0.008-0.512 mg/m L(r=0.9998); The average recovery rate was 100.31%, with a RSD value of 1.38%. The error of RSD in precision, stability and repeatability test were no more than 2%; the average content of paeoniflorin was 17.54 mg/g. Conclusion The method in this paper is simple, quick, accurate and reproducible, which can be used for controlling the content of paeoniflorin in Fufangshaohong Granules.
