胶原诱导性关节炎小鼠脾脏及踝关节中G蛋白耦联受体激酶2的表达

摘  要:目的 :了解胶原诱导性关节炎(collagen-induced arthritis,CIA)发病后免疫器官及免疫细胞内G蛋白耦联受体激酶2(G protein-coupled receptor kinase 2,GRK2)的表达及其变化,探讨其参与CIA发病的机制。方法:用鸡Ⅱ型胶原(collagen typeⅡ,CⅡ)免疫DBA/1J小鼠;免疫荧光双标法检测小鼠踝关节和脾脏中CD4、Foxp3、ROR-γt分别和GRK2的共定位;Western Blot法检测正常小鼠和CIA小鼠踝关节和脾脏中GRK2的表达变化。结果 :CIA小鼠四肢踝关节明显红肿;正常小鼠与CIA小鼠脾脏组织中均有CD4、Foxp3、ROR-γt和GRK2的共定位;CIA小鼠踝关节中也有CD4、Foxp3、ROR-γt和GRK2的共定位;CIA小鼠脾脏和踝关节组织中的GRK2的表达均降低。结论 :CIA小鼠脾脏及踝关节组织表达GRK2且GRK2的表达显著低于正常小鼠。Objective: To investigate the expressions and changes of G protein-coupled receptor kinase 2(GRK2) in immune organs and immune cells from collagen-induced arthritis(CIA) mice, and to explore the pathogenesis of GRK2 involved in the of CIA. Methods: CIA was induced by chicken type Ⅱ collagen(C Ⅱ ) in DBA/1J; Colocalization of CD4, Foxp3, or ROR-γt, and GRK2 from ankle and spleen in normal and CIA mice were detected by double immunofluorescent staining respectively; GRK2 expression in ankle and spleen from normal and CIA mice were determined by Western Blot. Results: Significant swelling of ankle joint was observed in CIA models, colocalization of CD4, Foxp3, ROR-γt and GRK2 in spleen from both normal and CIA mice were observed respectively; colocalization of CD4, Foxp3, ROR-γt and GRK2 were also detected in ankle tissue from CIA mice models; during the inflammatory process, the expression of GRK2 was downregulated from the tissue of spleen and ankle joint of CIA. Conclusions: GRK2 was expressed in spleen and ankle joint tissues from CIA and normal mice, the level of GRK2 expression was profoundly decreased in CIA compared with normal mice.
