大黄总蒽醌胶囊的血清药理学研究

摘　　要：目的:建立大黄总蒽醌胶囊血清药理学的研究方法。方法:以细胞增殖力和谷草转氨酶(aspartate transaminase,AST)渗漏为指标建立过氧化氢(hydrogen peroxide,H2O2)诱导的HL-7702肝细胞损伤模型,并考察体系中添加不同浓度大黄总蒽醌胶囊含药血清对损伤肝细胞的影响。结果:0.5 mmol/L和2.5 mmol/L H2O2分别为细胞增殖试验和AST渗露检测的适宜造模浓度;体系中添加5%大黄总蒽醌胶囊含药血清损伤肝细胞增殖明显增加(P<0.05),添加10%、20%、40%含药血清损伤肝细胞增殖明显降低(P<0.01或P<0.05);体系中添加5%和10%含药血清损伤肝细胞AST渗漏明显减少(P<0.01),添加20%、40%含药血清损伤细胞AST渗漏明显增加(P<0.01)。结论 :大黄总蒽醌胶囊含药血清具有肝细胞保护和毒性双向作用,低剂量对损伤肝细胞具有保护作用,高剂量则产生毒性。Objective： To establish a serum pharmacology method for total rhubarb anthraquinone capsules（TRAC）. Methods： Taking cell proliferation and AST in cell culture as pharmacological indices, the model of H202-induced injury of HL-7702 was established, and the effects of different addition amount of drug-containing serum in culture system on injury hepatocytes were evaluated. Results： The appropriate concentration of injury model were 0.5 and 2.5 mmol/L, respectively. The former was determined for cell proliferation test and the later was determined for AST infiltrated detection. Addition of 5% drug- contained serum to culture system could significantly elevate cell proliferation（P〈0.05） and addition of 10%, 20%, 40% drug- contained serum eould signifieantly decrease cell proliferation（P〈0.01 or P〈0.05）; addition of 5%, 10% drug-contained serum to culture system could significantly reduce AST infiltrated concentration（P〈0.01） and Addition of 20%, 40% drug-contained serum could significantly increase AST infiltrated concentration（P〈0.01）. Conclusion： The serum containing drug of TRAC had obviously bidirectional effect on liver. Low dose of addition had protective effect on injured hepatocytes, while high dose of addition had toxic effect.
