摘  要:目的探讨4 w自愿转轮运动对慢性不可预知应激致抑郁大鼠下丘脑乙酰胆碱代谢及下丘脑室旁核c-fos表达的影响。方法随机将30只大鼠分为对照组(C,n=10)、应激模型组(SM,n=10)及运动应激组(SEX,n=10)。SEX及SM组大鼠每日进行慢性不可预知性应激1次,连续28 d,SEX组大鼠同时进行4 w自愿转轮运动。慢性应激及转轮运动结束后检测大鼠下丘脑乙酰胆碱酯酶(Ach E)及胆碱乙酰基转移酶Ch AT的活性,采用免疫组织化学结合图像半定量方法对下丘脑室旁核c-fos神经元的数量、面积及灰度进行测量和分析。结果与C组比较,慢性应激大鼠下丘脑Ach E及Ch AT活性均显著增强(P<0.01,P<0.01),下丘脑室旁核c-fos表达显著增加(P<0.01);经过4 w自愿转轮运动,与SM组大鼠比较,SEX大鼠下丘脑Ach E及Ch AT活性显著减弱(P<0.05,P<0.05),下丘脑室旁核c-fos表达降低(P<0.05)。结论长期自愿转轮运动可以纠正慢性应激致抑郁模型大鼠下丘脑胆碱能系统功能亢进并减弱下丘脑室旁核c-fos的表达。Objective To explore the influence of the 4-week voluntary wheeling exercise on the acetylcholine metabolism within hypothalamus and c-fos expression within paraventricular nucleus( PVN) of the hypothalamus in the depressive rats induced by chronic unpredictable stress( CUMS). Methods 30 Sprague Dawley male rats were randomly divided into the control( C,n = 10),the stress model( SM,n = 10) and stress exercise groups( SEX,n = 10). The rats of SEX and SM groups received CUMS every day and lasted 28 days. At the same time,rats in SEX group received 4-week voluntary wheeling exercise. Then Ach E and Ch AT activities within hypothalamus were detected and c-fos expression within PVN of the hypothalamus was detected and analyzed by ABC immunohistochemical technique and semi quantitative.Results Compared with C group,Ach E and Ch AT activities were increased in hypothalamus and c-fos expression was enhanced in hypothalamus PVN in the SM group( P〈0. 01). Compared with SM group,Ach E and Ch AT activities were reduced( P〈0. 05) in hypothalamus and c-fos expression was decreased( P〈0. 01) in hypothalamus PVN in the SEX group. Conclusions Voluntary wheeling exercise could rectify the acetylcholine hyperfunction within hypothalamus and weaken c-fos expression within hypothalamus PVN in depressive model rats.
