摘  要:为了研究咖啡酸锗对小鼠U14腹水瘤的抑制作用及作用机制,试验建立小鼠U14腹水瘤模型,观察咖啡酸锗对U14腹水瘤小鼠体质量及生命延长率的影响,MGG染色观察U14瘤细胞的形态变化,FCM检测U14瘤细胞的凋亡率,并观察细胞周期。结果表明:咖啡酸锗能改善U14腹水瘤小鼠的生存状态,提高生命延长率,以中剂量效果最佳;MGG染色可见较多凋亡的肿瘤细胞;咖啡酸锗能诱导U14肿瘤细胞凋亡,在G0-G1前出现一个明显的亚二倍体凋亡峰,使细胞周期被阻滞在S期。说明咖啡酸锗能改善荷瘤小鼠体质量,提高生命延长率,并能诱导肿瘤细胞凋亡,这可能是其发挥抗肿瘤作用的机制之一。To study the inhibitory effect and active mechanism of caffeic acid germanium (Ge) on the U14 ascitic tumor in mice, U14 ascitic tumor model in mice was established to observe the effect of eaffeic acid Ge on the body mass of and the life extension rate of mice with UI4 ascitic tumor. Morphologic change of U14 tumor cells was observed using the May - Gr/inwald - Giemsa (MGG) staining and the apoptosis rate of U14 tumor cells was detected using FCM, while the cell cycle was also observed. The results showed that eaffeic acid germanium could improve survival condition and increase life extension rate of mice with U14 aseitic tumor, and it was found that the effect of medium dose of caffe- ic acid Ge was best. More apoptosis cells were seen through MGG staining, and FCM showed that caffeic acid germanium could induce the apoptosis of tumor U14 cells. Hypodiploid apoptosis peak appeared in the GO -G1 phase, which made the cell cycle blocked in the S phase. The results indicate that carrie acid germanium can improve body mass of tumor - beating mice, and improve the life extension rate, and induce ap- optosis of tumor ceils, which is one of mechanisms for the anti - cancer effect of caffeic acid Ge.
