摘 要:为了探讨甘露(聚)糖结合凝集素(Mannan-binding lectin,MBL)基因在绵羊不同组织和品种间的表达差异,试验利用实时荧光定量PCR技术分析MBL基因在绵羊8个不同组织器官和外周血白细胞中的表达差异,以及在4个品种中的表达差异。结果表明:MBL基因在绵羊心脏、体侧部皮肤、肝脏、肺脏、肾脏、小肠、脾脏、小脑及血液白细胞中具有不同程度的表达,其中肝脏中表达量最高,肺脏和脾脏中表达量次之,心脏和小肠中表达量最低。MBL基因在哈萨克羊肝脏中表达量最高,中国美利奴羊(新疆军垦型)肝脏中表达量次之,萨福克羊和湖羊肝脏中表达量最低。说明MBL基因具有组织特异性表达特征,并且在不同绵羊品种中表达存在差异,其表达量与绵羊的适应性有关。To investigate expression differences of mannan -binding lectin (MBL) gene in different tissues; different breeds of sheep, a real - time fluorescent quantitative PCR assay was used to analyze the expression differences of MBL genes in eight different tissues, organs and peripheral blood leukocytes in sheep, as well as in four breeds of sheep. The results showed that MBL gene had different levels of expression in the sheep heart, body side skin, liver, lung, kidney, small intestine, spleen, cerebellum and white blood ceils. The expression level of MBL gene in liver was highest, followed by lung and spleen, but the lowest expression levels were in heart and Small intestine. The expression level of MBL gene was highest in liver of Kazakh sheep, followed by China Merino from Xinjiang Military Reclamation, but the lowest expression levels were in the livers of Suffolk and Hu sheep. The results indicate that MBL gene has tissue - specific expression feature, and there are differences in breeds of sheep, while the expression level of MBL gene is related to the adaptation of sheep.
