摘 要:为了解促红细胞生成素受体(EPOR)对动物低氧适应的重要作用,试验选取饲养在西藏林芝(海拔约3 000 m)的高原藏猪、高原大约克猪和饲养在北京(海拔约100 m)的大约克猪(低地大约克猪),在6月龄进行屠宰,采集心脏、肝脏和肺脏器官组织,使用荧光定量PCR技术测定EPOR基因的表达量。结果表明:猪EPOR基因在肺脏器官组织中表达量比较高,在心脏和肝脏器官组织中表达量较低。高原藏猪EPOR基因的表达量在3个器官组织中均高于高原大约克猪和低地大约克猪,但在肺脏器官组织中高原藏猪与高原大约克猪相比差异不显著(P>0.05)。大约克猪从低地移居到高原地区,器官组织中EPOR基因表达量均显著增加(P<0.05)。EPOR基因是受低氧调控的基因,低地大约克猪移居到高原之后该基因表达量增加;高原藏猪EPOR基因表达量高于大约克猪,可能对其高原适应性具有重要作用。To understand the important role of erythropoietin receptor (EPOR) in hypoxic adaptation of animals, heart, three groups of pig population were selected in the test, including highland Tibetan and Yorkshire pigs raised at LinZhi of Tibet at an altitude of about 3 000 - meter altitudes and Yorkshire pigs raised in Beijing at an altitude of about 100 - meter altitudes. All pigs were slaughtered at six months of age, and the heart, liver and lung tissues were collected. The EPOR gene expression was 'determined using real - time fluorescence quantitative PCR technique. The results showed that the expression level of the EPOR gene in the lung tissue was higher than that in the heart and liver organs and tissues among three pig groups. The expression levels of EPOR gene in the three tissues of highland Tibetan pigs were higher than those of highland Yorkshire pigs and lowland Yorkshire pigs, but there was significant difference ( P 〉 0.05 ) in the expression level of EPOR gene in the lung tissues and organs between highland Tibetan pigs and highland Yorkshire pigs. When Yorkshire pigs were moved from the lowland to the plateau region, the expression levels of EPOR in the the organs and tissues were significantly increased (P 〈 0.05 ). The resuhs indicate that the EPOR gene is regulated by the hypoxia, and the EPOR expression level is increased in Yorkshire pigs after migrating to plateau. The EPOR expression level in highland Tibetan pigs is higher than that in Yorkshire pigs, which might be play an important role in its f plateau adaptability.
