摘  要:为了调查新疆部分地区规模化奶牛场致犊牛腹泻轮状病毒的感染情况,查明该病毒在致犊牛腹泻中的作用,试验采用双抗体夹心ELISA、RT-PCR的方法检测轮状病毒抗原及VP6基因,并对该基因进行序列分析以比较其同源性。结果表明:各牛场腹泻犊牛粪便的轮状病毒阳性检出率为23.08%-90.91%;在健康犊牛粪便中,仅从石河子某奶牛场检出轮状病毒,检出率为38.89%;RTPCR扩增得到大小为383bp的片段;8份克隆毒株序列的同源性为97.9%,同源性较高。说明新疆部分地区犊牛感染轮状病毒十分普遍,轮状病毒是引起犊牛腹泻的主要病原之一,部分健康犊牛带毒但未发病。To investigate the infection status of diarrhea caused by rotavirus in calves from large - scale dairy farms in partial region of Xinjiang, and identify its role in 'causing diarrhea in calves, the double - antibody sandwich ELISA and RT - PCR methods were used to detect rotavirus antigen and VP6 gene. Sequence analysis for the gene was performed to compare its homology. The results showed that the positive detection rates of rotavirus in feces from calves with diarrhea were from 23.08% to 90.91% in each dairy farm. Rotavirus was detected in healthy calf feces only from a cow dairy farm of Shihezi, and the positive detection rate was 38.89%. A fragment of 383 bp was obtained by RT - PCR amplification. The sequence homologies of eight Cloned strains were 97.99%, showing that they had higher homology. The result indicates that rotavirus infection in calves is very common, and rotavirus is one of the main pathogens causing diarrhea in calves, while partial healthy calves are infected but not sick.
