温肾益气颗粒对慢性阻塞性肺病大鼠模型肺组织炎症因子变化的影响研究

摘　　要：目的:研究温肾益气颗粒对慢性阻塞性肺病(COPD)大鼠模型肺组织炎症因子的变化。方法:将50只SD大鼠随机分为正常对照组、模型组、温肾益气颗粒高剂量组、温肾益气颗粒中剂量组、温肾益气颗粒低剂量组,每组10只,采用熏香烟加气管滴注脂多糖法建立COPD大鼠模型,观察大鼠日常状态,ELISA法测定大鼠肺组织匀浆液TNF-α、IL-8、IL-17A的含量,HE染色检查肺部病理改变。结果:与正常对照组相比,造模组大鼠的肺组织炎症因子含量明显升高;出现COPD症状,日常状态较差,肺组织病理变化明显;温肾益气颗粒高中剂量组能够减轻肺组织的病变程度,减轻炎症反应;与模型组相比,温肾益气颗粒高、中剂量组能明显降低肺组织TNF-α、IL-8、IL-17A的含量,日常活动与模型组相比较好;温肾益气颗粒低剂量组细胞因子与模型组相比,没有明显变化。结论:温肾益气颗粒能够降低大鼠肺组织中细胞因子的含量,减轻肺组织感染程度。Objective： To study the influence of Wenshen Yiqi Prescription on inflammatory cytokines of chronic obstructive pulmonary disease（ COPD） rats model. Method： Fifty female SPF rats were randomly divided into normal control group,model group,Wenshen Yiqi Prescription high dose group（ 8. 0 g·kg- 1）,Wenshen Yiqi Prescription medium dose group（ 4. 0 g·kg- 1） and Wenshen Yiqi Prescription low dose group（ 2. 0 g·kg- 1）,10 in each group. Smoked cigarettes and trachea drip lipopolysaccharides method is used to establish the COPD model. Observe the daily activities. By using ELISA we detected the levels of TNF- α,IL- 8 and IL- 17 A in thelung tissue. Lungtissue pathological changes were observed by HE staining. Result： Compared with the normal control group,others groups＇ TNF- α,IL- 8 and IL- 17 A levels wereincreased and the daily statuspoor as well as the obvious pathologicalchange of lung tissue. Compared with the model group,Wenshen Yiqi Prescription high dose group（ 8. 0 g·kg- 1） and medium dose group（ 4. 0 g·kg- 1） can reduce the levels of cytokines and alleviated the change of lung tissue. Wenshen Yiqi Prescription low dose group（ 2. 0 g·kg- 1） had no obvious change. Conclusion： Wenshen Yiqi Prescription can reduce the levels of TNF- α,IL- 8 and IL-17 A in the COPD rats and alleviate the pathological change.
