摘  要:目的:探究百合知母汤对慢性温和不可预见性应激(CUMS)结合孤养大鼠行为及单胺递质的影响;方法:50只SD大鼠随机分为5组,即正常对照组、模型组、氟西汀组、百合知母汤高剂量组、百合知母汤中剂量组,每组10只;除正常组外,其余4组动物给予慢性温和不可预见刺激结合孤养制造CUMS模型;从第4周开始,4组动物相应给予生理盐水、氟西汀、百合知母汤灌胃,持续4周;每组动物每周1次测定体重增长率,于第1、3、7周观测动物糖水偏好度、Open-field实验垂直与水平总得分;行为学指标测定完毕后,动物处死取血及大脑皮质,ELISA法测定5-HT、NE含量;结果:百合知母汤能增加大鼠体重增长率、糖水偏好度及Open-field实验垂直与水平总得分,增加血清及大脑皮质5-HT、NE的含量;结论:百合知母汤具有较好的抗抑郁效应,其机制与上调血清及大脑皮质中单胺递质水平相关。Objective: To study the effects of Baihe Zhimu Decoction on chronic mild unpredictable stress( CUMS)combined with isolated keeping depression rats' model behavior and monoamine neurotransmitters. Methods: A total of 50 SD rats were divided into 5 groups randomly,namely control group,model group,Fluoxetine group and high / moderate dosage of Baihe Zhimu decoction group. The depression model was made by using unpredictable chronic mild stimulation combined with depression isolated keeping to all rats expect rats of control group,in which the duration last for 3 weeks.From the beginning of 4 weeks,five groups' rats were given saline,fluoxetine,Baihe Decoction high / moderate dosage for4 weeks. Each rat was measured the body weight growth rate once a week during 7 weeks,also the rate of sucrose preference,vertical and horizontal total score in Open- field experiment in 1st,3rd,7th week respectively. All the animals were killed to get the blood and brain cortex to test the contents of 5- HT and NE by ELISA. Results: Baihe Zhimu Decoction can increase the body weight growth rate,sucrose preference degree and the Open- field experiment vertical and horizontal score,increase the contents of 5- HT and NE in serum and cerebral cortex. Conclusion: Baihe Zhimu Decoction has a better antidepressant effect,in which its mechanism is related to increasing the serum and the level of monoamine neurotransmitters in the cerebral cortex.
