摘 要:目的:比较西双版纳地区红杆铁皮石斛与青杆铁皮石斛茎上、中、下3个部位的多糖含量、甘露糖含量、甘露糖与葡萄糖峰面积比的差异。方法:采用苯酚-浓硫酸法测定多糖含量,柱前衍生化HPLC法测定甘露糖含量及甘露糖与葡萄糖峰面积比。结果:多糖含量,红杆铁皮石斛茎上、中部明显高于青杆铁皮石斛茎上、中部,而红杆铁皮石斛茎下部明显低于青杆铁皮石斛茎下部;甘露糖含量,红杆铁皮石斛上、中、下3个部位均明显高于青杆铁皮石斛;甘露糖与葡萄糖峰面积比,红杆铁皮石斛上、中、下3个部位均明显低于青杆铁皮石斛。红杆铁皮石斛的多糖含量、甘露糖含量及甘露糖与葡萄糖峰面积比均达到2015版《中华人民共和国药典》标准,青杆铁皮石斛仅多糖含量达到2015版《中华人民共和国药典》标准。结论:同一地区的红杆铁皮石斛与青杆铁皮石斛茎上、中、下3个部位主要有效成分含量有明显的差异,建议在临床用药过程中将红杆铁皮石斛与青杆铁皮石斛分开使用,同时将铁皮石斛按其茎上、中、下3个部位进行分级,为铁皮石斛种质资源品质评价及临床用药提供实验基础。Objective: To compare the contents of polysaccharide and mannose,the peak area ratio of mannose and glucose in different parts of red and green stems of Dendrobimn officinale Kimura et Migo in Xishuang Banna. Methods: In this method,Phenol- concentrated sulfuric acid method was used to determine the content of polysaccharide. HPLC with pre- column derivatization was used to determine the content of mannose and the peak area ratio of mannose and glucose.Results: The polysaccharide content of the upper and middle parts of the red stems of Dendrobimn officinale Kimura were significantly higher than those of the green stems and the lower parts of the red stems of Dendrobimn officinale Kimura were significantly lower than those of the green stems. The mannose content of the upper,middle and lower parts of the red stems of Dendrobimn officinale Kimura were significantly higher than those of the green stems. The peak area ratio of mannose and glucose of the upper,middle and lower parts of the red stems of Dendrobimn officinale Kimura were significantly higher than those of the green stems. The content of polysaccharide and mannose,the peak area ratio of mannose and glucose in different parts of the red stems of Dendrobimn officinale Kimura et Migo reached to the standard of Chinese Pharmacopoeia 2015. The content of polysaccharide of the green stems of Dendrobimn officinale Kimura et Migoreaches reached to the standard of Chinese Pharmacopoeia 2015. Conclusion: The main active ingredient content in different parts of the red and green stems of Dendrobimn officinale Kimura et Migo in same area have significant differences,and so the suggestion was given to using the red and green stems of Dendrobimn officinale Kimura et Migo separately in clinical medicine and classifing Dendrobimn officinale Kimura et Migo accroding to the upper,middle and lower parts of its stems,providing the experimental basis for the quality evaluation of germplasm resources and clinical medication of Dendrobimn officinale Kimura et Migo.
