摘  要:目的:探讨米屈肼对大鼠心肌氨基酸代谢的影响及其意义。方法:36只SD雄性大鼠随机分为对照组、米屈肼低剂量(100 mg·kg^(-1)·d^(-1),M100)和高剂量(300 mg·kg^(-1)·d^(-1),M300)组,每组12只。连续灌胃15 d后,开胸取心脏;Waters AccQ·Tag法测定3组心肌游离氨基酸浓度。结果:与对照组比,M100组中组氨酸(His)、精氨酸(Arg)、苏氨酸(Thr)、丙氨酸水平下降(P<0.05);M300组中谷氨酸(Glu)、His、Arg水平下降,天冬氨酸(Asp)、甘氨酸、Thr、脯氨酸、半胱氨酸、酪氨酸、甲硫氨酸(Met)、赖氨酸(Lys)水平上升(P<0.05)。随着米屈肼给药剂量提高,Asp、Met、Lys水平逐渐上升,Glu水平逐渐下降。结论:米屈肼能影响大鼠心肌氨基酸代谢。Objective: To investigate the potential effect of mildronate on myocardial amino acid metabolism in rats. Methods: Thirty-six male SD rats were randomly divided into control group, mildronate doses of 100 mg·kg-1·d-1 (M100) and 300mg·kg-1·d-1 (M300)groups, 12 each group. After all rats were treated for 15 days by gastric lavage, their hearts were removed and the concentration of myocardial free amino acid of rats was determined by Waters AccQ'Tag. Results: In the M100 group, the concentration of histidine (His), argnine (Arg), threonine (Thr) and alanine decreased comparing to control group (P〈0.05). In the M300 group, the concentration of glutamate (Glu), His, Arg decreased and the concentration of aspartate (Asp), glycine, Thr, proline, cysteine, tyrosine, methionine (Met), lysine (Lys) increased comparing to control group (P〈0.05). As the dose of mildronate was increased, the concentration of Asp, Met, Lys increased gradually and the concentration of Glu decreased gradually. Conclusion: Mildronate can affect myocardial amino acid metabolism in rats.
