摘 要:采用SRAP分子标记对32个杜鹃花品种进行了分析。21对引物共检测到232个多态位点,平均每对引物扩增出12.2个位点,多态位点百分率为90.27%。32个杜鹃花品种的Nei's基因多样性指数(H)为0.340,Shannon's信息指数(I)为0.503,相似系数介于0.541~0.805之间。3个品种群间遗传分化系数(G_(st))为0.157 2,表明84.28%遗传变异发生在品种群内。采用UPGMA法构建系统进化树,32个杜鹃花品种可分为3类:14个东鹃品种、10个西鹃品种和8个夏鹃品种,结果与传统分类完全一致。表明SRAP分子标记技术能准确地分析杜鹃花品种的遗传多样性和亲缘关系。Genetic diversity across 32 varieties of Rhododendron simiss was analyzed using sequence-related amplified polymorphism(SRAP).A total of 232 polymorphic bands were generated using 21 pairs of SRAP primers and the polymorphism percentage was 90.27%.Each pair of primers generated 12.2on average.Nei's gene diversity was 0.340,Shannon's information index was 0.503 and the similarity coefficient varied from 0.541 to 0.805.In addition,the gene differentiation coefficient(Gst)was 0.157 2,which showed that84.28% genetic diversity existed within the populations.The 32 varieties could be divided into three groups based on the result of UPGMA:14varieties of"Dongjuan"population,10 varieties of"Xijuan"population and 8varieties of"Xiajuan"population,which was consistent with the traditional classification completely.These results indicated that artificial breeding causes confusion in the genetic background of R.simsii varieties.However,SRAP could be applied to study the genetic diversity and relationship of R.simsii accurately.
