摘  要:本研究旨在通过分析与脂肪性状相关的A-FABP和H-FABP基因单核苷酸多态性,找出优势基因型个体即保持胡须鸡的肉质口感又能克服生长速度慢的特点,为下一步的基因型选择提供理论基础。本实验以惠阳胡须鸡作为实验材料,从鸡血液中提取DNA,通过PCR扩增、测序等分子生物学技术分析了A-FABP和H-FABP基因单核苷酸多态性。结果表明:1)A-FABP基因发现了两处碱基突变,一个在第1685碱基处由C突变T,但由于不在编码区,没有氨基酸的改变,而另一个在第1830碱基处缺失了G,引起编码氨基酸的改变即缺失了D(天冬酰胺),进而导致蛋白质空间结构发生改变。2)H-FABP基因发现两个碱基突变,其中一个突变位点不在编码区,没有相应氨基酸改变,而另一个在第746碱基处由A突变为G,使得编码的氨基酸由赖氨酸变成谷氨酸,导致蛋白质空间二级结构发生改变。这些结果说明在惠阳胡须鸡中,A-FABP和H-FABP基因存在单核苷酸多态性,为下一步的基因型选择奠定了理论依据。This study aims to analyze single nucleotide polymorphisms (SNP) of A-FABP and H-FABP genes which are associated withfat traits in order to find out the advantageous genotypes that maintain good-quality meat but also fast-growth for genotype selection infuture. In this experiment, Huiyang bearded chickens were used as experimental material. DNA was extracted from the chickens' blood,followed by PCR amplification and sequencing, then single nucleotide polymorphisms of A-FABP and H-FABP genes were analyzed.Results show that: 1 ) two base mutations were found in A-FABP gene: one in 1685 from C to T does not change the amino acid becauseit is not located in the coding region, but another in 1830 missing G, alter encoded amino acid that results in deleting D (asparagine),which leads to the change of spatial structure of A-FABP; 2) two base mutations were found in H-FABP gene: one of them is not in thecoding region, so there is no corresponding amino acid changes, and another in 746 from A to G results in lysine into glutamic, whichchanges the spatial structure of H-FABP. These results indicate that in Huiyang chickens, there exist single nucleotide polymorphismsof A-FABP and H-FABP genes, which will lay the theoretical foundation for genotype selection in the future.
