摘 要:为研究甘蓝根肿病菌的生物学特性和在寄主根部的侵染过程,以湖北长阳火烧坪根肿病菌为试验材料,研究不同温度、pH及培养基质对甘蓝根肿病菌休眠孢子萌发的影响,并利用光学显微镜初步观察该病原菌在寄主根部的侵染情况。结果显示,在温度24℃、pH 6.5以及过滤除菌的根分泌液条件下,休眠孢子的萌发率最高。通过观察根肿病菌不同时间段的侵染情况发现,第3天时游动孢子开始侵染根毛;第5天时形成大量原质团,且根毛顶端开始膨大;第8天时大量的原质团开始分化;第11~13天时形成游动孢子囊,且部分游动孢子成熟释放;第25天时观察到根部肿大,且镜检发现根皮层细胞内充满大量的休眠孢子囊。说明甘蓝根肿病菌与温度、pH以及培养基质密切相关,且侵染过程是一个实时动态变化的过程。In order to elucidate biological characteristics and infection progress of Plasmodiophora brassicae,the clubroot were collected from Changyang country,Hubei province. The germination of resting spores was studied under different temperature,pH and culture substrate conditions,and infection process was observed by optical microscope. The optimum temperature for the germination of resting spores of P.brassicae was 24 ℃,the optimum pH was 6. 5 and root exudates were suitable for the germination of resting spores. The infection in the host root was observed under different time points. The results showed that the root hair was infected after inoculation 3 days,primary plasmodia were found in the root hair at 5 days and more swollen root hair tips were observed,the plasmodia began to divide gradually at 8 days,and then zoosporangia was formed and the number of zoosporangia was increased from 11 to 13 days. Small root galls were observed after inoculation 25 days,with root cortical cells full of the resting spores. Therefore,Plasmodiophora brassicae was closely related to temperature,pH and the root exudates,and root hair infection was a dynamic process.
