摘 要:目的探究龟板对多发性抽动症(tourette syndrome,TS)模型大鼠的行为学改变,及其对纹状体多巴胺(dopamine,DA)、多巴胺D_2受体(dopamine D_2receptor,D_2R)、血浆中DA的含量的影响。方法将40只大鼠分为空白组、模型组、硫必利组和龟板组。腹腔注射亚氨基二丙腈(iminodipropionitrile,IDPN)建立TS模型,并给予每组相应的生理盐水或药物灌胃。造模完成后每周进行行为学检测,并于造模第4周采用ELISA法检测纹状体DA、D_2R、血浆中DA的含量。结果 (1)与模型组比较,两周后硫必利组大鼠刻板行为减少(P<0.05),三周后运动行为减少(P<0.05),四周后运动行为明显减少(P<0.01)。与模型组比较,龟板组两周后大鼠刻板行为与运动行为均减少(P<0.05);(2)模型组大鼠较空白组大鼠血浆中DA含量升高(P<0.05),纹状体DA含量降低(P<0.05),纹状体D_2R含量升高(P<0.05)。与模型组比较,龟板组大鼠血浆DA含量减少(P<0.05),硫必利组血浆DA明显减少(P<0.01),纹状体DA含量两组均升高(P<0.05),两组纹状体D2R未发生明显变化(P>0.05)。结论龟板对TS有良好的治疗作用,其原因可能与调节DA的代谢有关。Objective To investigate the behavior change of Plastrum Testudinis on Tourette's syndrome( TS) in a rat model,and the influence of Plastrum Testudinis on striatal dopamine( DA),dopamine D_2receptor( D_2R) and the content of DA in plasma,and provide neural biochemical basis of Plastrum Testudinis in treatment of TS. Methods 40 rats were divided into blank group,model group,Tiapride group and Plastrum Testudinis group. Rats were injected intraperitoneally with iminodipropionitrile( IDPN) to establish the TS model,and the rats weregiven corresponding normal saline and drugs. Behavioral testing was performed weekly after the model was completed,and the contents of DA,D_2 R and DA in the striatum were detected by ELISA method at fourth weeks after the model was made. Results( 1) Compared with model group,stereotyped behavior of Tiapride group was reduced( P0. 05) after 2weeks,motor behavior was reduced after 3weeks( P0. 05),motor behavior was reduced obviously after 4weeks( P0.01). Compared with model group,stereotyped behavior and motor behavior of Plastrum Testudinis group was reduced( P0. 05).( 2) Compared with blank group,DA in plasma of model group was increased( P0. 05); DA in corpus striatum was reduced( P0. 05) and D_2 R in corpus striatum was increased( P0.05). Compared with model group,DA in plasma was reduced in Plastrum Testudinis group( P0. 05) and significantly reduced in Tiapride group( P0. 01),DA in corpus striatum were all increased in two group( P0. 05),D_2 R in corpus striatum did not change obviously( P0. 05). Conclusions The influence of Plastrum Testudinis on rat model of TS was significantly,and the reason maybe related to the regulation of DA metabolism.
