摘  要:目的观察冬虫夏草菌粉对大鼠肾缺血-再灌注损伤动物模型Klotho基因和免疫调节的影响,探讨其可能的机制。方法 30只Wistar大鼠分为假手术组、模型组、实验组。模型组和实验组大鼠采用夹闭双肾动脉45 min后复灌方法造模,假手术组不夹闭双肾动脉作为对照。术后实验组给予10%冬虫夏草菌粉按2.0 m L/(kg·d)灌胃,假手术组和模型组则给予2.0 m L/(kg·d)生理盐水。灌胃治疗7 d后,检测尿磷(UP)含量、血清总蛋白(STP)、白蛋白(ALB)和肾功能3项[胱抑素C(Cys C)、肌酐(Scr)和尿素氮(BUN)],RT-PCR法检测肾组织Klotho蛋白和Klotho-m RNA表达及肾组织匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)和乳酸脱氢酶(LDH)含量。结果模型组肾组织匀浆中Klotho蛋白和Klotho-m RNA与假手术组比较下调(均P<0.05);模型组大鼠心室内血STP和ALB治疗后与假手术组比较差别不大(均P>0.05)。实验组Klotho蛋白、Klotho-m RNA、STP和ALB与模型组比较均升高(均P<0.05)。模型组心室内血清SOD与假手术组比较降低,而MDA升高(均P<0.05),LDH则差别不大(P>0.05)。实验组MDA与模型组比较降低,SOD活性增强(均P<0.05)。模型组Cys C、Scr、BUN和UP与假手术组比较均升高(均P<0.05)。实验组Cys C、Scr、BUN和UP与模型组比较均降低(均P<0.05)。结论冬虫夏草菌粉在抑制肾缺血-再灌注造成的脂质过氧化反应的同时,免疫力得以提升,并通过上调Klotho蛋白和lotho-m RNA达到抗衰老、增加UP排泄来减轻肾功能损害。Objective: To establish the rat renal ischemia - reperfusion injury animal model, to observe the effect of Cordyceps sinensis on the Klotho gene and immune regulation of the anti-ischemia-reperfusion injury and immune regulation mechanism. Methods: 30 Wistar rats were randomly divided into sham operation group,the model group and the experimental group. The model group and the experimental group were modelled with reperfusion method 45rain after clamping bilateral renal, sham operation group without clamping bilateral renal as con- trol. After operation,the experimental group received 10% Cordyceps powder, with garage of 2.0 mL/(kg, d), sham operation group and the model group with 2.0 mL/(kg, d) irrigation saline as control. 7 days after gavage administration, urine phosphorus (up) content was detected;heart blood samples were taken for measurement of STP, ALB and renal function in three [Cys C,SCRand BUN]. Rats were sacrificed from the right kidney and detected through RT-PCR to detect Klotho protein and Klotho-mRNA expression,then the content of MDA,SOD and LDH in renal tissue. Results: Compared with the model group,STP,ALB and SOD in the experimental group increased; Klotho protein and Klotho-mRNA significantly up-regulated ; Cys C, SCR, BUN and UP decreased, and MDA was significantly inhibited(P〈 0.05). Compared with sham operation group, Klotho protein and Klotho-mR- NA significantly down-regulated in the model group (all P〈 0.05);there was little difference between the two groups in STP and ALB after treatment (all P 〉 0.05). Compared with the model group,Klotho protein,Klotho- mRNA, STP and ALB in the experimental group rose (all P〈 0.05). Compared with the sham operation group, the serum SOD in the model group was lower,while the MDA was increased (all P〈 0.05).
