摘 要:目的探讨大鼠脑创伤(TBI)骨髓间充质干细胞(BMSCs)超顺磁性氧化铁颗粒(SPIO)标记及活体磁共振成像(MRI)示踪的可行性。方法参照Feeney法制作大鼠左侧TBI模型,右侧大脑立体定向注射移植SPIO标记的BMSCs。实验组大鼠注射移植SPIO标记BMSCs,对照组TBI大鼠分别注射等量的无SPIO标记的BMSCs和PBS,均分别于注射细胞前及注射BMSCs后1、3、7、14和21d进行NSS评分及MRI扫描,观察脑实质内信号变化及其迁移情况,并与对侧正常脑实质信号进行对比。每次MRI扫描结束后立即各处死1只,取脑组织切片进行HE及普鲁士蓝染色。结果 MRI扫描观察到实验组TBI模型大鼠脑实质内低信号区,并逐步向病变区迁移,与对侧脑实质内信号减低率比较具有统计学意义(F=154.19,P<0.05),病理切片普鲁士蓝染色见蓝染颗粒。BMSCs移植组NSS评分低于PBS移植组,结果有统计学意义(P<0.05)。结论 MRI对大鼠TBI模型立体定向注射SPIO标记的BMSCs活体示踪有一定价值,BMSCs对TBI大鼠神经功能恢复具有一定作用。Objective To evaluate the feasibility of tracking transplanted SPIO-labeled bone marrow mesenchymal stem cells with magnetic resonance imaging in traumatic rats.Methods TBI model was established in the left hemisphere of the rats.The rats were randomized into 3 groups that received injection of either SPIO-labeled BMSCs in the right hemisphere,non-SPIO-labeled BMSCs or PBS only.All rats were analyzed by NSS and scanned with MRI pre-implantation,1 day,3 days,1 week,2 weeks and 3 weeks after implantation.Signal changes and cell immigration in brain tissue were observed and compared with the contralateral sphere.After each MRI scanning,brain tissue was subject to HE and Prussian blue staining.Result In rats injected with SPIO-labeled BMSCs,significant lower signal intensity was observed in injected sphere compared with the contralateral sphere on MRI with the scanning sequences of FSE T2WI and Truefisp T2*,F =154.19,P 0.05).Cell immigration towards the lesions was observed.Blue particles were observed in brain sections after prussian blue staining.Rats receving BMSC transplantation had lower NSS than PBS group(P 0.05).Conclusion MRI may be used to track transplanted SPIO-labeled BMSCs in rat TBI model.Transplantation of BMSC may promote recovery of neural function.
