摘 要:目的观察干扰素-α治疗猪血清诱导大鼠肝纤维化的作用,并探讨其治疗肝纤维化的作用机制。方法 60只Wistar雄性大鼠完全随机分为3组,每组20只,分别为正常组、猪血清模型组和IFN-α治疗组。正常组给予0.9%生理盐水腹腔注射,猪血清模型组和IFN-α治疗组给予猪血清腹腔注射,每次0.5 m L/只,2次/周,共12周。自第9周第1天开始,正常组和猪血清模型组给予0.5%CMC Na灌胃,每天1次,共4周;IFN-α治疗组给予IFN-α皮下注射10×104U/只/天,共4周。于12周末处死所有大鼠,留取肝脏组织,行Masson染色进行肝纤维化组织学半定量评分;化学比色法检测肝组织内羟脯氨酸含量。结果猪血清模型组肝纤维化组织学半定量评分及羟脯氨酸表达量均高于正常组(P<0.01),IFN-α治疗组较猪血清模型组分别降低32.8%和32.5%(P<0.01)。结论 IFN-α对猪血清诱导的大鼠肝纤维化具有治疗作用,其作用机制可能与减少肝脏组织中胶原含量有关。Objective: Observing and discussing the operating mechanism of interferon-αto cure rat hepatic fibrosis induced by pig serum. Methods: Sixty male Wistar rats are divided into randomly three groups( n= 20 in each group): normal control group,pig serum model group, and IFN-αgroup. The rats in normal control group are injected physiological saline into the abdominal cavity(0.5 ml each time, twice a week for 12 weeks). From the 9th week, the rats for the normal group and model group are given0.5% CMC Na into stomach(once a day for 4 weeks). 10×104U subcutaneous injection for the IFN-αgroup(each one a day for 4weeks). All rats are killed but left liver at the end of the 12 th week. Pathological changes and semi-quantitative score of the liver are made by Masson staining, Hydroxyproline content in liver tissue are detected in chemical colorimetric method. Results: the semi-quantitative score for hepatic fibrosis and the contents of hydroxyproline for the model group are higher than that in the normal group(P0.01), and these are increased by 32.8% and 32.5% respectively. Conclusion: the treat function of the liver induced by pig serum, and its mechanism for IFN-α may have relation with reducing the contents of collagen in liver.
