摘  要:[目的]用5种不同方法制取牡丹叶下表皮制片并对其气孔进行比较研究。[方法]采用直接撕皮法、撕皮-刮去叶肉细胞法、NaOH离析法、过氧化氢-冰醋酸离析法和煮沸撕取法制取牡丹叶下表皮,对其制片指标进行了比较,并筛选出最佳染色剂。[结果]撕皮-刮去法和过氧化氢-冰醋酸离析法的制片效果较好,叶肉残留少、真实性强。0.5%的I-KI染液染色效果好。[结论]该研究结果可为今后不同品种牡丹叶表皮的微形态特征研究奠定基础。[Objective] Five different methods of Paeonia suffruticosa leaf lower epidermis slice-preparing and the observation of stomata were comparatively studied. [Method] The slice of Paeonia suffruticosa leaf lower epidermises were prepared with the method of directly tearing leaf epidermis,tearing leaf epidermis-scrape off mesophyll cells,NaOH maceration,H_2O_2-HAC maceration and boiling-tearing leaf epidermis and the preparation index was compared and the best stain was screened. [Results] The treatment of tearing leaf epidermis-scrape off mesophyll cells and H_2O_2-HAC maceration was best in all of preparation methods,which was less residual mesophyll and highly facticity. In the dyeing treatment,the treatment of 0. 5% I-KI showed the good efficiency. [Conclusion] The foundation for the future research on the micro-morphological characteristics of Paeonia suffruticosa leaf lower epidermis of different varieties would be laid.
