摘 要:目的:利用在体多通道电生理学记录技术研究前扣带皮层吻侧部(rostral anterior cingulate cortex,rACC)神经元参与情绪情感活动时的放电特征时,除在rACC脑区埋置多通道电极外,还需同时埋置给药管以观察药物对rACC神经元放电特征的影响。本文将介绍带给药管的多通道在体记录电极的制作及埋置方法。方法:自制带双侧给药管的多通道在体记录电极,所采用给药管材质有三种,分别为金属、表面无涂层的石英玻璃纤维(玻璃裸管)和表面有聚酰胺胶涂层且具一定柔韧性的石英玻璃纤维(玻璃涂层管),然后埋植电极并记录神经电信号,最后插管位置组织定位。结果:盐水被注射到目标脑区,即rACC脑区;电极丝也定位于双侧前扣带皮层吻侧部;用带金属给药管的多通道电极,记录到的神经元放电背景噪音较大(信噪比<3)。用带玻璃裸管的电极可清晰记录到目标神经元的放电活动(信噪比>3),信噪比与金属管组比较[(5.37±1.35)vs(2.35±0.64)],差异有统计学意义(P<0.05),但该玻璃管质硬易碎,不易插入堵丝,也不易在慢性实验中脑部长时间保存。用带玻璃涂层管的电极也可清晰记录到神经元的放电活动(信噪比>3),信噪比与金属管组比较[(5.20±1.21)vs(2.35±0.64)],差异有统计学意义(P<0.05),且易于插入堵丝,便于在动物脑中长时间保留。结论:自制带涂层石英毛细玻璃管的电极成功实现了对某一脑区或核团同步埋植给药管和电极,使得药物干预并记录清醒活动大鼠脑电活动的实验便于进行。Objective:In order to explore the firing characteristics of the rostral anterior cingulate cortex (rACC) neurons,which participate in the emotional activities by using the multi-channel in vivo recording techniques,we should implant a multi-channel electrode and cannulas in the rACC area in rats,while to observe the influence of drugs on the firing characteristics in ACC neurons at the same time.A method of home-made multi-channel with double cannulas is introduced here.Method:Three different materials of cannulas,including metal, quartz capillary without or with polyamide coating were applied.Then the home-made electrode was implanted to record the neuronal activity.In the end,the cannula site was localized.Result:The saline were respectively injected into ACC region in different rats,and electrode wire are also located in this region.Using home-made multi-channel electrode with double metal cannulas,the signal to noise ratio were smaller (SNR〈3).Electral signal could be recorded by using the home-made multi-channel electrode with double quartz capillary without or with polyamide coating, their signals both were excellent (SNR〉3),the SNR compared with metal cannulas one[(5.37±1.35) vs (2.35±0.64)],there had significant statistical difference(P〈0.05),but the bare quartz capillary was harder,fragile and difficult to maintain for a long time in the brain.With glass coating tube electrode could also be clearly recorded neurons discharge activity(SNR〉3),the SNR compared with metal cannulas one[(5.20±1.21) vs (2.35±0.64)],there had significant statistical difference(P〈0.05),while the coating one was elastic and easy to insert a plug in it and can maintain a long time.Conclusion:The home-made multi-channel electrodes with double cannulas can be successfully applied to some of experiments,which need to be recorded about neural activities after drugs administration to one brain area or neural nuclei.
