摘  要:目的:运用高效液相色谱法(HPLC)测定右美托咪定丙泊酚混合注射液的水相浓度,探讨右美托咪定影响丙泊酚注射痛的作用机制。方法:将待测的丙泊酚注射液按右美托咪定的浓度分为三组,每组5例样本。其中A组为单纯丙泊酚注射液组,B组丙泊酚注射液含右美托咪定0.75μg/mL,C组丙泊酚注射液含右美托咪定1.25μg/mL。反透析法得到游离在水相的丙泊酚后,用高效液相色谱仪测定各样本丙泊酚水相游离浓度,比较三组注射液水相浓度。结果:A组丙泊酚水相浓度最高,C组最低,三组水相浓度比较差异均有统计学意义(P<0.05)。结论:右美托咪定可能通过降低丙泊酚水相浓度影响丙泊酚注射痛的发生。Objective:To use high performance liquid chromatography (HPLC)to determinate the Dexmedetomidine mixed with Propofol injection aqueous phase concentration and investigate the effect of Dexmedetomidine mechanism of Propofol injection pain.Method:Propofol injection will be tested according to the concentration of Dexmedetomidine were divided into three group,each group had 5 cases.Group A was simply Propofol injection group,group B was given Propofol injection with Dexmedetomidine 0.75μg/mL,group C was given Propofol injection with Dexmedetomidine 1.25μg/mL.The aqueous phase free concentration of Propofol in each sample was determined by HPLC after the reverse dialysis method used in the water phase and the aqueous phase concentration of the three groups were compared.Result:The highest aqueous phase concentration was group A,the lowest aqueous phase concentration was group C,there were significant differences between three groups in the aqueous phase concentration(P〈0.05).Conclusion:Dexmedetomidine alleviates the propofol injection pain likely through decreasing the concentration of Propofol in aqueous phase of injection solution.
