摘 要:目的:探讨大肠癌细胞株HM7、HT29间辐射敏感性差异与共济失调毛细血管扩张症突变(ATM)基因的关系。方法:应用半定量反转录聚合酶链反应(RT-PCR)方法,分析10Gy X线照射前后HM7、HT29中ATM mRNA表达水平变化。结果:在X线照射前,HT29中ATM mRNA水平比HM7高;在照射后,HM7细胞的ATM mRNA水平在早期(1h)就出现轻度上升(是基础水平的1.4倍);而HT29细胞在照射后4h才表现为ATM mRNA量上升(是基础水平的1.6倍):两者在照射后12h左右ATM mRNA表达水平恢复接近基础水平。结论:经X线照射后,在对放射相对敏感的HT29与对放射相对抗拒的HM7细胞中,ATM mRNA水平变化规律存在差异。这种ATM mRNA表达的不同可能是导致两者放射敏感性不同的原因之一。Objective:To investigate the relationship between ataxia-telangiectasia mutant(ATM)gene and the different radiosensitivity of two nasopharyngeal colorectal cell lines,that is HM7 and HT29.Methods:Semi-quantitive reverse transcription polymerase chain reaction(RT-PCR)was used to measure the level of ATM mRNA in HM7 and HT29when exposed to 10 Gy X-rays irradiation.Results:Without irradiation,the basic ATM mRNA levels in HT29 were slightly lower than in HM7.After 1hour of irradiation,the ATM mRNA levels of HT29 increased to 1.6times than basic levels.But the ATM mRNA levels of HM7 increased to 1.4times than basic levels after 4hours of irradiation respectively.At last,returned to basic levels at 12 hours after irradiation.Conclusion:After X-ray irradiation,in the cells of HT29 which is sensitive to radiation and HM7 with radiation resistance,their variation of ATM mRNA levels was different.it might be one of the reasons of different radiosensitivity.
