摘  要:目的了解系统性红斑狼疮(SLE)病人体内Epstein-Barr(EB)病毒潜伏膜蛋白1(LMP-1)基因、Ⅰ型干扰素诱导基因(ISGs)表达的关系及其与SLE的相关性。方法选择SLE病人68例(其中狼疮肾炎病人39例)、健康查体者(正常对照组)40例,采用实时荧光定量PCR方法检测两组外周血淋巴细胞中EB病毒LMP-1基因及ISGs mRNA表达水平,并分析二者相关性。结果 SLE组EB病毒LMP-1mRNA水平显著高于正常对照组(U=1 039.0,P〈0.05);SLE病人LMP-1 mRNA表达与SLE疾病活动度积分呈正相关(r=0.22,P〈0.05)。SLE病人ISGs中的2′-5′寡腺甘酸样合成酶(OASL)、干扰素刺激基因15(ISG15)和淋巴细胞抗原6复合体E(LY6E)mRNA表达水平高于正常对照组(U=377.5~725.0,P〈0.05),黏液病毒抗性蛋白1(MX1)mRNA水平两组间比较差异无显著性(P〉0.05)。肾炎组与非肾炎组ISG15mRNA表达水平差异有显著性(U=387.5,P〈0.05)。SLE病人EB病毒LMP-1与MX1、OASL、ISG15基因mRNA水平呈正相关(r=0.003~0.219,P〈0.05),而与LY6EmRNA无显著相关(P〉0.05)。结论 EB病毒LMP-1基因可能通过激活Ⅰ型干扰素通路导致SLE的发病与病情进展。Objective To investigate the correlation between expression of latent membrane protein 1(LMP-1)and typeⅠinterferon induced genes(ISGs)of Epstein-Barr(EB)virus in patients with systemic lupus erythematosus(SLE). Methods A total of 68 patients with SLE-including 39 with lupus nephritis-and 40 healthy controls were enrolled in this study.Real time reverse transcription polymerase chain reaction(RT-PCR)was used to test the mRNA levels of LMP-1and ISGs,and the correlation between the two expressions was analyzed. Results In SLE group,the level of LMP-1mRNA was higher than that in the control group(U=1 039.0,P〈0.05),the expression of LMP-1mRNA in SLE patients was positively correlated with the disease activity score(r=0.22,P〈0.05).In SLE group,the expression levels of OASL,ISG15 and LY6Ewere higher than that in the normal control group(U=377.5-725.0,P〈0.05),while the difference in MX1 mRNA level between the two groups was not significant(P〉0.05).The difference in ISG15 mRNA expression between patients with lupus nephritis and without was significant(U=387.5,P〈0.05).The mRNA level of LMP-1was positively correlated with MX1,OASL and ISG15 mRNA level(r=0.003-0.219,P〈0.05),but was not significantly correlated with LY6EmRNA(P〉0.05). Conclusion EB virus LMP-1is likely by activating the type-1interferon pathway leading to the development and progression of systemic lupus erythematosus.
