摘 要:目的建立香港海鸥形菌(Laribacer hongkongensis)的基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)鉴定方法,并对其实验室应用加以评价。方法用VITEK MS全自动快速微生物质谱检测系统对12株香港海鸥形菌进行鉴定和分析,用SARAMIS数据库构建参考图谱;并用新分离的香港海鸥形菌、嗜水气单胞菌、温和气单胞菌、溶血不动杆菌、琼氏不动杆菌、腐败希瓦氏菌、藤黄微球菌、类志贺邻单胞菌等表型相似的菌株对所建立的鉴定方法进行验证。结果本实验构建的香港海鸥形菌参考图谱特异性高、重复性好,是细菌鉴定数据库中未见的新图谱;新分离香港海鸥形菌菌株均能被MS成功鉴定,且图谱吻合率在98.5%以上,其他25株表型相似菌株均能被成功鉴别。结论MALDI-TOF-MS技术能快速和准确地鉴定香港海鸥形菌,此方法可用于香港海鸥形菌的临床快速诊断或食品安全性评价。Objective To establish the identification method for Laribacter hongkongensis( LH) by matrix-assited laser desorption ionization-time of flight mass spectrometry( MALDI-TOF-MS),and to evaluate its applications in laboratory. Methods Identification and analysis of 12 strains of LH were performed by VITEK MS automatic rapid microbial mass spectrometry detection system,and the reference spectrum was constructed with SARAMIS database; this MS identification method was validated by several newly isolated strains( Laribacter hongkongensis,Aeromonas hydrophila,Aeromonas sobria,Acinetobacter haemolytius,Acinetobacter johnsonii,Shewanella putrefaciens,Micrococcus luteus,Plesiomonas shigelloides) which have similar phenotypes with LH. Results LH reference spectra established in this study had high specificity and reproducibility,which was a new spectra for the strain identification database. The newly isolated LH strains could be identified successfully with the coincidence rate more than 98. 5%,and the other 25 strains which had similar phenotypes with LH can be discriminated successfully. Conclusion MALDI-TOF-MS could be a rapid and accurate technology for the identification of Laribacter hongkongensis,and may have applications in both clinical rapid diagnosis and food safety detection.
