摘  要:目的:观察天麻素联合电针对局灶性脑缺血大鼠额叶皮质胶质纤维酸性蛋白(glial fibrilally acidic protein,GFAP)和神经元特异性烯醇化酶(neuron-specific enolase,NSE)表达的影响,以期探讨针药结合对局灶性脑缺血治疗的机制。方法:SD大鼠50只随机分为正常、模型、天麻素、电针及天麻素+电针5个组别。采用线栓法复制局灶性脑缺血大鼠模型。天麻素组按10 mg/kg剂量腹腔注射天麻素注射液,每天1次,共治疗14d;电针组电针刺激"百会"和"足三里"穴,刺激30 min,每天1次,共治疗14 d;天麻素联合电针组给予天麻素与电针治疗,每天1次,共治疗14d。免疫组织化学方法结合图像分析系统检测脑缺血侧额叶皮质GFAP和NSE的表达。结果:与正常组比较,模型组GFAP的表达增强,NSE的表达减少,均有统计学差异(P<0.05)。与模型组相比较,天麻素组、电针组和天麻联合电针素组GFAP的表达明显减少,NSE的表达明显增加,均有统计学差异(P<0.05)。天麻联合电针素组与天麻素组或电针组比较,GFAP的表达减少,NSE的表达增加,也有统计学差异(P<0.05)。结论:天麻素与电针均可能抑制内源性神经干细胞(endogenous neural stem cells,e NSCs)向神经胶质细胞分化,而促进e NSCs向神经元分化,二者结合的调节作用更显著,这可能是天麻素联合电针具有较好抗脑缺血损伤的作用机理之一。AIM:To observe the effect of electroacupuncture (EA) combined with gastrodin on the expression of glial fibrilally acidic protein (GFAP) and neuron specific enolase (NSE) in the dentate gyms (DG) of focal cerebral ischemia(FCI) rats,so as to explore its mechanism underlying improvement of differentiation at Hippocampal eNSCs of FCI.METHODS:Fifty Sprague-Dawley rats were randomly divided into the normal,model,EA,gastrodin and EA + gastrodin groups.FCI rats were induced by middle cerebral artery occlusion (MCAO) with thread embolus.The gastrodin group was injected with a daily 10 mg/kg dose of gastrodin intraperitoneally for 14 days.The EA group was given daily 30 min electro-acupuncture on left side “ Baihui ”(DU20) and “Zusanli” (ST36) for 14 days.The EA + gastrodin group was given a combined treatment of gastrodin and electro-acupuncture for 14 days.The expressions of GFAP and NSE in DG were detected by immunohistochemical and image analysis system.RESULTS:Compared with the normal group,the GFAP expression level of the model group was significantly increased (P 〈 0.05),while the NSE expression level was significantly decreased (P 〈 0.05).Compared with the model group,the GFAP expression levels were down-regulated in the EA,gastrodin and EA + gastrodin group (P 〈0.05),while the NSE expression levels were decreased.Compared with the gastrodin or the EA team,the GFAP expression level of the EA + gastrodin team was decreased while the NSE expression level was increased (P 〈 0.05).CONCLUSION:Both EA and gastrodin can significantly promoted eNSCs differentiation into neurons instead of glial cells.The effect is more obvious in the EA + gastrodin group,which may be one of the possible mechanisms underlying its good resistance to cerebral ischemia injury.
