摘  要:目的:石榴复合果醋发酵过程中,还原糖的含量是衡量发酵过程正常与否的重要标志.建立既符合保健果醋特点,又能简便、快速的还原糖测定方法,具有重要的科学意义.方法:建立3,5-二硝基水杨酸比色定糖法(DNS法)、直接滴定法、萨氏法三种还原糖测定方法,比较三种方法的精密度和准确度,优选最佳测定方法;同时,对最优方法的精密度、检测限、定量限和加标回收率进行分析,考察脱色与否对石榴复合果醋还原糖测定的影响,确定最优测定条件检测石榴复合果醋中还原糖的含量.结果:1 3,5-二硝基水杨酸比色定糖法、直接滴定法、萨氏法的RSD分别为:0.0049%、0.0060%、0.0330%;加标回收率分别为:100.41%、105.56%、97.35%,确定DNS法的精密度和准确度均优于其他两种方法,更适用于还原糖的测定,既简便又快速;2通过对精密度、检测限、定量限和加标回收率的比较分析,色素对还原糖的测定结果影响较大,将样品利用活性炭脱色处理,可提高DNS法测定的精确性;3石榴复合果醋中还原糖的最终含量为50.92g/L.结论:DNS法重现性好,方法简便、快捷,适用于石榴复合果醋发酵过程中还原糖的测定.Objective:In the fermentation process of pomegranate compound health fruit vinegar,the content of reducing sugar is an important indicator concerning whether the fermentation process is normal or not. It is of significance to establish a simple and fast method to analyze the reducing sugar in the health fruit vinegar. Methods:Three experimental methods including the the 3,5-Dinitrosalicylic acid (DNS) method, direct titration, and somgyis method for determination of reducing sugar were compared in terms of the precision and accuracy in order to select the optimal determination method. To optimize the protocol, the analysis of the precision, limit of detection, limit of quantitation, recovery rate was carried out to identify the influence of decoloration on the determination of reducing sugar of the pomegranate compound fruit vinegar. Results: ①RSD of the DNS method, direct titration and somgyis method were 0. 0049% ,0. 0060% and 0. 0330% ,respectively,with the standard addition recovery of 100. 41% , 105.56% and 97. 35% , indicating that the precision and accuracy of DNS method were better than those of the other two methods and thus more suitable for simple and fast determination of the reducing sugar;②The analysis on the precision, limit of detection, limit of quantitation and recovery rate showed that the pigment exhibited great impact on the detection result of reducing sugar. Decoloration of samples with the activated carbon could greatly improve the precision of the DNS test; ③The content of the reducing sugar in the pomegranate compound vinegar was 50. 92g/1. Conclusion:DNS method is simple, fast and has good reproducibility, suitable for the determination of reducing sugar during the fermentation process of the pomegranate compound fruit vinegar.
