摘 要:以带有玉米褪绿斑驳病毒(MCMV)的玉米叶片为材料,研究建立了MCMV的普通RT-PCR、TaqMan实时荧光RT—PCR和SYBRGreenI实时荧光RT-PCR检测方法,并比较了3种方法的灵敏度。结果表明:TaqMan实时荧光RT-PCR的检测灵敏度最高.最低检出量可达1.61fg,SYBRGreenI实时荧光RT—PCR略逊之,普通RT—PCR的检测灵敏度则相对较低.与前两者相差10-100倍。RT-PCR, TaqMan real-time RT-PCR and SYBR Green I real-time RT-PCR approaches were developed for detection of Maize chlorotic mottle virus(MCMV) , and their sensitivity in detection of MCMV was compared. Results showed that the sensitivities of TaqMan real-time RT-PCR and SYBR Green I real-time RT-PCR methods were ten to one hundred times higher than that of conventional RT-PCR, in particular the TaqMan real-time RT-PCR, which displays the highest sensitivity with a detectable limit of 1.61 fg.
