丁酰胆碱酯酶抑制剂对急性高血脂小鼠模型血脂的影响

摘　　要：目的观察丁酰胆碱酯酶抑制剂对急性高血脂小鼠血脂水平的影响。方法采用腹腔注射泰洛沙泊（400mg／kg）的方法建立小鼠急性高血脂模型，并通过灌胃给药的方式给予辛伐他汀（10mg／kg）、丁酰胆碱酯酶特异性抑制剂四异丙基焦磷酸亚胺（ISO-OMPA）（1mg／kg）和非特异性抑制剂新斯的明（0．5mg／kg），分别分析丁酰胆碱酯酶被抑制的程度以及小鼠血浆中血脂的变化。结果新斯的明对丁酰胆碱酯酶的体外抑制效果是ISO—OMPA的136倍，而ISO—OMPA体内抑制效果强于新斯的明；ISO—OMPA和新斯的明均可显著抑制小鼠血清丁酰胆碱酯酶的活性，模型组小鼠的血浆总胆固醇（TC）、甘油三酯（TG）和低密度脂蛋白胆固醇（LDL—C）浓度分别为（586．1±99．8）、（277．2±92．7）、（164．6±82．5）mg／dl，新斯的明组与之比较分别下降了19％、15％和6％。结论丁酰胆碱酯酶与血脂明显相关。Objective To study the influence of butyrylcholinesterase （BuChE） inhibitors on the serum lipids of acute hyperlipidemia model. Methods Tyloxapol （400 mg/kg） was introperitoneally injected to build the mouse acute hyperlipidemia model, and the mice were administrated with simvastatin （ 10 mg/kg）, four isopropyl imine pyrophosphate （ ISO - OMPA, 1 mg/kg） and neostigmine （0. 5 mg/kg） by intragastric administration method. The inhibitory degree of activity of BuChE and the serum lipid was analysized by enzyme linked immunosorbent assay （ELISA）. Results The inhibitory effect of neostigmine was better than ISO - OMPA in vitro, but the inhibitory effect of ISO - OMPA was better than neostigmine in vivo. Both ISO - OMPA and neostigmine could inhibit the activity of BuChE with statistically significant difference. The concentrations of serum total cholesterol （TC）, triglyceride （TG） and low density lipoprotein cholesterol （ LDL - C ） in model group were （ 586. 1 ± 99. 8 ）, （ 277. 2 ± 92. 7 ）, and （ 164. 6 ± 82. 5） mg/dl respectively, the concentrations of serum TC, TG and LDL - C in neostigmine group were reduced by 19%, 15% and 6% respectively as compared with model group. Conclusion The activity of BuChE has a significant correlation with serum lipids.
