摘  要:目的:采用LC-HR-MS/MS技术研究紫草中酚酸类化学成分。方法:采用LC-HR-MS/MS法,使用C18(4.6 mm×100 mm,1.8μm)色谱柱,以乙腈-0.1%甲酸溶液为流动相,梯度洗脱,流速0.4 m L·min^(-1),柱温30℃,进样量10μL;质谱条件:HESI离子源;正负离子扫描模式;雾化温度为300℃;离子传输管温度为380℃,质量扫描范围为m/z 100~1 500。结果:以酚酸类化合物的质谱裂解规律为指导,参照紫草中已报道成分的质谱数据,分析推断了紫草中可能含有紫草酸、咖啡酸四聚体、迷迭香酸、和丹酚酸C等9个化学成分。结论:本实验建立的分析方法能够较全面地反映出紫草中酚酸类的化学成分,为进一步开发利用该药材提供实验依据。Objective: To study the chemical constituents of the phenolic acids in Arnebia euchroma(Royle) Johnstn.by the method of LC-HR-MS/MS.Methods: The HPLC separation was performed on a C18 column(4.6 mm×100 mm, 1.8 μm) in gradient elute mode with acetonitrile and water(containing 0.1% formic acid) at a flow rate of 0.4 m L·min(-1), the column temperature was 30 ℃, and the injection volume was 10 μL.HESI was employed, the sample was analyzed in the positive and negative-ion mode, with the probe heater temperature at 300 ℃,capillary temperature 380 ℃, and m/z range was from 100 to 1 500.Results: Phenolic acids compounds were identified by mass spectrometry(MS) as lithospermic acid, caffeic acid tetramer, rosmarinic acid and salvianolic acid C etc.Conclusion: The established LC-MS/MS analysis method can reflect the phenolic acids compounds from Arnebia euchroma(Royle) Johnstn.comprehensively, and thus provides an experimental basis for further exploitation.
