衍生气相色谱法测定鸡疫苗中游离甲醛的含量

摘　　要：建立了鸡疫苗中游离甲醛含量测定的衍生气相色谱法。用2,4-二硝基苯肼衍生鸡疫苗中的游离甲醛后,环己烷萃取衍生物,HP-5毛细管色谱柱分离,气相色谱分析,电子捕获检测器检测,外标法定量。游离甲醛质量浓度在0.05-5 mg/L内与峰面积呈良好的线性关系,回归方程为Y=2781.9X＋14.525（r=0.9999）。平均加样回收率为99.44%（RSD=1.87%）。本方法准确稳定,灵敏度高,重复性好,可用于鸡疫苗中游离甲醛的含量测定。A GC method for the determination of free formaldehyde in poultry vaccine has been developed. The sam ples were derivatized by 2,4-dinitrophenylhydrazine,extracted by cyclohexane,purifid with H P-5 capillary colum n,seperated by GC with an electron capture detector and qualified by external standard calibration curves. The free form aldehyde in the vaccine showed a good linear relationship to peak area within a concentration range of 0. 0 5 - 5 m g / L,the regression equation was：Y = 2 7 8 1. 9 X ＋ 1 4. 5 2 5（ r = 0. 9 9 9 9）. The average recovery using the established m ethod was9 9. 4 4 %（ RSD = 1. 8 7 %）. The derivatization capillary gas chrom atography m ethod is developed,which is accurate, stable, sensitive, reproducible and m ight be used for determ ination of free form aldehyde in poultry vaccine.
