摘 要:目的本实验主要探讨孕前砷暴露对F1代新生鼠睾丸发育和vasa基因表达的影响。方法将24只雌性SD大鼠随机分为3组(n=8),通过灌胃方式建立砷染毒模型后,"一代一窝"繁殖试验技术获得后代。统计新生鼠的数量并称重,镜下观察睾丸组织结构并测量生精索直径及索间面积,实时荧光定量PCR和免疫组织化学法检测新生鼠睾丸组织vasa mRNA和vasa蛋白表达。结果与对照组(0.9%Na Cl)比较,高剂量组(1.5 mg/kg/d As_2O_3)新生鼠的出生数、活胎率和新生鼠平均体重都在显著降低,有统计学差异(P<0.05);与对照组比较,中剂量组(0.75 mg/kg/d As_2O_3)生精索直径变窄和索间面积变宽,有统计学差异(P<0.05),高剂量组生精索直径变窄和索间面积变宽程度加剧,差异极显著(P<0.01);与对照组比较,中剂量组、高剂量组的vasa mRNA和vasa蛋白表达量显著下降(P<0.01);与中剂量组比较,高剂量组vasa mRNA表达量明显下降(P<0.05),vasa蛋白表达量显著下降(P<0.01)。结论随孕前染砷剂量的增加,F1代新生鼠生精索直径变窄,索间面积增大,vasa mRNA和vasa蛋白表达量降低。因此,孕前砷暴露对新生鼠睾丸发育产生一定抑制作用。Objective This experiment mainly discusses the effect of neonatal rats testis development and vasa gene expression after pregestational arsenic exposure.Methods The 24 female SD rats were randomly divided into three groups(n = 8).Establishment of arsenic exposure model by means of gastric lavage,offspring were obtained through " One generation nest" breeding test technology.Counted the number and weight of newborn rats,Observed the structure of the testis tissue and measured the diameter of the cord and the area between the cords.The expression of vasa mRNA and vasa protein were detected by RT-PCR and immunohistochemistry.Results Comparison of high dose group(1.5 mg/kg/d As2O3) and control group(0.9% Na Cl),there were significant differences(P〈0.05) in the number and average weight and live birth rate of newborn rats.Compared with the control group,the diameter of the cord narrows and the area between the cords broaden,the medium dose group(0.75 mg/kg/d As2O3) was statistical difference(P〈0.05),the high dose group was significantly different(P〈0.01);Compared with the control group,the expression of vasa mRNA and vasa protein decreased significantly(P〈0.01).Compared with the medium group,the vasa mRNA expression of the high dose group decreased significantly(P〈0.05),and the expression of vasa protein decreased significantly(P〈0.01).Conclusions With increasing doses after pregestational arsenic exposure,the diameter of the cord were narrows,the area between the cords were increases,the expression of vasa mRNA and vasa protein decreased.After pregestational arsenic exposure,neonatal rat testis development has a certain inhibitory effect.
