摘  要:目的初探贵州省产金钗石斛总碱(DNLA)对APP/PS1转基因小鼠空间学习记忆的影响。方法雄性APP/PS1转基因小鼠随机分为模型对照组、模型给药组(DNLA,40 mg/kg),同月龄野生型小鼠作为空白对照组,每组10只。自7月龄始灌胃给药,连续8周。Morris水迷宫及Y迷宫检测小鼠空间学习记忆能力,尼氏染色观察小鼠海马神经元形态。结果Morris水迷宫定位航行实验显示,模型组逃避潜伏期较空白对照组明显延长(P<0.05);空间探索实验显示,模型组小鼠在原平台象限停留时间及在原平台象限游泳距离的百分比较空白组明显缩短(P<0.05);模型组小鼠在Y迷宫的空间自发性探索行为正确率明显低于空白组。尼氏染色结果显示,模型组小鼠海马神经元排列疏松,神经元数量及尼氏小体减少,胞质呈淡蓝色。与模型组比较,DNLA 40 mg/kg明显缩短小鼠逃避潜伏期(P<0.05);增加小鼠在原平台象限停留时间及原平台象限游泳距离的百分率(P<0.05)、明显增加Y迷宫小鼠自发探索行为正确率(P<0.05),并改善模型小鼠海马CA1区神经元的形态和结构。结论 DNLA对APP/PS1转基因小鼠空间学习记忆能力具有改善作用。Objective To explore the effects of Dendrobium nobile Lindl.alkaloids(DNLA) on the spatial learning and memory function of APP/PS1 transgenic mice.Methods Male APP/PS1 transgenic mice were randomly divided into model and DNLA(40 mg/kg) groups,age-matched wildtype mice as controls ten mice in each group.Administration was performed from 7 months of age,and continued for 8 weeks.Morris water maze and Y maze were used to test the spatial learning and memory abilities.Nissl's staining was used to observe the morphological changes of hippocampus neurons.Results Place navigation tests of Morris water maze showed that the escape latency was longer in model group than control group(P〈0.05),while the probe trial assay data demonstrated that the time and the percentage of distance in the target quadrant were decreased in model group(P〈0.05).The percentage of spontaneous alternation behavior correction in model group was decreased.The numbers of neurons and Nissl's bodies were decreased in model group by Nissl's staining,Compared with model group,DNLA 40 mg/kg reduced the escape latency(P〈0.05),increased the time and the percentage of distance in the target quadrant(P〈0.05),increased the percentage of spontaneous alternation behavior correct rates(P〈0.05),and improved the morphology and structure in CA1 area of hippocampus.Conclusion DNLA has the ability of improving the spatial learning and memory function of APP/PS1 mice.
