黄连水提物对小鼠白色念珠菌性阴道炎的影响

摘　　要：目的探讨黄连水提物对小鼠白色念珠菌性阴道炎的治疗作用。方法采用微量肉汤稀释法测定黄连水提物对白色念珠菌的最小抑菌浓度（minimum inhibitory concentration,MIC）,通过病理组织切片观察黄连水提物对阴道黏膜刺激的病理变化;构建小鼠白色念珠菌性阴道炎模型,对黄连水提物治疗的模型小鼠阴道内的白色念珠菌菌落进行计数,观察黄连水提物对阴道组织及模型小鼠阴道炎病程的影响。结果黄连水提物对白色念珠菌有较好的体外抑菌活性,MIC为0.94～3.75 mg/m L;病理切片结果显示,与空白组比较,20～80 mg/m L的黄连水提物对小鼠阴道黏膜无明显局部刺激,100 mg/m L黄连水提物对小鼠局部黏膜刺激明显;与阴性对照组比较,高、中剂量的黄连水提物能明显减少模型小鼠的阴道分泌物菌落数（P〈0.01）,且可明显缩短模型小鼠的病程（P〈0.01）;低剂量的小鼠阴道内菌落数减少较明显（P〈0.05）,病程缩短较明显（P〈0.05）。结论黄连水提物对白色念珠菌有较强的体外抑菌活性,对白色念珠菌性阴道炎模型小鼠有良好的治疗作用。Objective To investigate the therapeutic effect of Coptis chinesis aqueous extract against mice model of candidal vaginitis.Methods Broth microdilution to measure the minimum inhibitory concentration（MIC） of Candidal albicans with Coptis chinesis.Adopting the pathological tissue sections to observe the pathological changes of the stimulation of vaginal mucosa under the influence of Coptis chinesis.We establish the mouse model of candidal vaginitis to detect the effect of Coptis chinesis on the colony number in vaginal mucosa of mice and observe the effect of Coptis chinesis on vaginal tissue and the course of disease.Results Coptis chinesis aqueous extract had a good antibacterial activity against Candida albicans,MICs were 0.94 ～ 3.75 mg/ml;Compared with the blank group,pathological section results showed that 20 ～ 80 mg/ml Coptis chinesis aqueous extract had no obviously local stimulation in mice vaginal mucosa,but 100 mg/ml Coptis chinesis aqueous extract stimulate the mice＇s vaginal mucosa obviously.Compared with the negative group,the high and medium dose of Coptis chinesis aqueous extract could significantly reduce CFU of Candida albicans vaginitis in mice secretion（P〈0.01）,and could significantly shorten the course of disease.Low dose Coptis chinesis aqueous extract can shorten the disease course（P〈0.05） and reduce the number of vagina colonies obviously（P〈0.05）.Conclusion Coptis chinesis aqueous extract has good antibacterial activity against Candida albicans in vitro,and a good therapeutic effect against candidal vaginitis.
