摘  要:目的观察人参茎叶总皂苷(TG)对博莱霉素(BLM)致小鼠肺间质纤维化模型的保护作用。方法将120只昆明种小鼠随机分为假手术对照组、模型对照组、人参茎叶总皂苷(40、80、160 mg/kg)组及醋酸泼尼松(5 mg/kg)组。气管内灌注博来霉素(5 mg/kg)建立小鼠肺纤维化模型,制模后连续给药28 d,观察各组小鼠肺组织病理形态改变,检测小鼠血清中丙二醛(MDA)、谷胱甘肽(GSH)含量,总抗氧化能力(T-AOC)、羟自由基(.OH)水平,超氧化物歧化酶(SOD)及髓过氧化物酶(MPO)活力。结果制模28 d后,模型组小鼠肺系数明显高于假手术对照组(P<0.01),肺泡炎及肺纤维化明显,血清中.OH、MPO水平及MDA的含量增高,GSH含量、T-AOC水平及SOD的活力显著下降(P<0.05)。与模型对照组比较,TG(80、160 mg/kg)及醋酸泼尼松给药能使模型小鼠的肺系数明显降低(P<0.01);肺纤维化程度明显减轻,且血清中.OH水平、MPO活力、MDA含量显著下降,而使GSH含量、T-AOC水平及SOD活力明显提高,差异具有统计学意义(P<0.05)。结论 TG对博来霉素所致的小鼠肺纤维化具有保护作用,其作用机制可能与其抑制氧化损伤有关。Objective To observe the protective effect of total ginsenosides on bleomycin(BLM) induced differentiation of mouse model with pulmonary fibrosis.Methods 120 Kunming mice(bisexual each half) are randomly divided into Sham-operation control group,model control group,total ginsenosides,TG(40,80,160 mg/kg) and prednisone group(5 mg/kg).After molding continuous administration 28 days,we will observe the pathological changes of lung tissue in every group of mice and superoxide dismutase(SOD),malon-dialdehyde(MDA),glutathione(GSH),total antioxidant capacity(T-AOC),hydroxyl radical(.OH),myeloperoxidase(MPO) vitality in serum of mice were detected.Results After molding 28 days,the lung coefficient of model mice is significantly higher than sham group(P〈0.01),alveolitis and pulmonary fibrosis become obvious,the content of.OH,MPO,MDA in serum significantly increase(P〈0.05),GSH content,T-AOC level and activity of SOD was remarkably decreased(P〈0.05).The administration with TG and prednisone to make mice lung index decrease and pathomorphism ameliorate significantly comparing with the model group(P〈0.01).Conclusion TG induced protection against pulmonary fibrosis by bleomycin in mice are involved,at least in part,to its inhibitory effects on oxidative damage.
