摘  要:目的 探讨促性腺激素释放激素激动剂(gonadotropin releasing hormone agonist,GnRHa)预处理对性成熟雌鼠调节性T细胞(regulatory T cells,Treg)的影响。方法 30只动情周期正常的7-8周未交配KM雌鼠随机分成5组,第1组连续9 d每日注射醋酸曲普瑞林1次,第2组连续9d每日注射醋酸曲普瑞林后,在第9天注射重组人促卵泡激素1次,第10天注射绒毛膜促性腺激素1次,第3组、4组分别与第1组、2组对应,用生理盐水代替醋酸曲普瑞林,其余处理不变,第5组无药物干预。流式细胞学检测各组外周血、脾脏T淋巴细胞亚群及Treg比例;并分别从前4组中随机选取4只小鼠留取子宫内膜,用实时荧光定量PCR方法检测子宫内膜Foxp3 mRNA含量。结果 流式细胞技术检测结果发现,外周血T淋巴细胞亚群及Treg比例各组比较无明显差异。比较脾脏T淋巴细胞亚群及Treg比例发现,与第5组比较,第1、2、3、4组CD3+比例下降(P〈0.05),第2组CD4^+比例下降(P〈0.05);第2组CD4^+CD25^+Foxp3^+Treg比例高于第4、5组(P〈0.05);CD8^+比例在各组间未见明显差异。实时荧光定量PCR检测结果发现,与第3组比较,第1组子宫内膜Foxp3 mRNA升高(P〈0.05);与第4组比较,第2组子宫内膜Foxp3 mRNA升高(P〈0.05)。结论 GnRHa预处理后促排卵,可提高脾脏Treg比例及子宫内膜Foxp3 mRNA含量,可能提高机体免疫耐受水平。This study designed to investigate the effects of gonadotropin-releasing hormone agonist (GnRHa) pretreatment on regulatory T cells (Treg) in mature female mice. Total of 30 unmated (7-8 weeks old) KM female mice with normal estrous cycle were randomly divided into 5 groups: group 1 received triptorelin acetate injection once daily for 9 consecutive days; group 2 also received triptorelin acetate injection once daily for 9 consecutive days, then received recombinant human follicle-stimulating hormone (r-hFSH) and choriogonadotropin (HCG) on the 9 th and 10 th days respectively; group 3 and 4 were corresponded with those of groups 1 and 2 respectively, but used saline instead of triptorelin acetate; group 5 was normal control with no drug intervention. Flow cytometry technology was used to evaluate the T cell subpopulation and Treg in peripheral blood and spleen of each group. RT-PCR was implemented to detect the expression of Foxp3 mRNA in the endometrium. Data showed that there was no significant difference in the proportion of T cell subpopulation and Treg in peripheral blood among the groups. But, the proportion of CD3^+T cell in the group 5 was lower than that of the other four groups (P〈0.05); the proportion of CD4^+T cell in group 2 were lower than that of the group 5 (P〈0.05);the proportion of CD4^+CD25^+Foxp3^+T cell in group 2 was higher than that of the group 4 and group 5(P〈0.05). No significant difference in proportion of CD8^+T lymphocyte was found among the groups. Furthermore, the expression of Foxp3 mRNA in the endometrium of the group 1 was higher than that of the group 3 (P〈0.05), and the expression of Foxp3 mRNA in the endometrium of the group 2 was higher than that of the group 4(P〈0.05). In conclusion, stimulating ovarian with the pretreatment of GnRHa could increase the proportion of CD4^+CD25^+Foxp3^+T cell in spleen and the expression of Foxp3 mRNA in endometrium, and then upregulates the function of immune tolerance.
