摘 要:骨桥蛋白(osteopontin,OPN),又称早期T淋巴细胞激活因子-1,具有经典的分泌型OPN(sOPN)和新近被证实的胞内型骨桥蛋白(iOPN)两种亚型,在免疫反应的不同层面中扮演重要角色.OPN能够促进免疫介导疾病和炎性疾病进展、调节宿主对感染的反应.OPN的作用及机制在部分疾病中已被阐明,但在多种疾病中仍不明确.OPN与一系列免疫相关疾病相关,包括多发性硬化、阿尔茨海默病、系统性红斑狼疮及B细胞相关性疾病等.值得关注的是,OPN与致病性T细胞尤其是Th17细胞关系密切;OPN对B细胞分化的作用也逐渐清晰.OPN一方面在上述疾病中具有致病性,但某些情况下又具有保护作用,因此未来需要更多研究阐明OPN在不同疾病中的作用及机制.Osteopontin,also known as early T lymphocyte activating factor-1,includes classically secreted form and newly identified intracellular form,playing important roles in the regulation of the immune response on multiple levels.It participates in the development of immune-mediated and inflammatory diseases,and regulates the host responses to infection.The effects and mechanisms have been elucidated in some diseases,but are still not clear in more cases.In this review,we focus on the role of osteopontin in a series of immune-mediated diseases,including multiple sclerosis,Alzheimer' s disease,Systemic Lupus Erythematosus and B Cell-related Diseases.Notably,osteopontin is closely related to pathogenic T cells,especially T helper 17 cells.The role osteopontin played in B-cell differentiation is becoming clear.On the one hand,osteopontin contributes to the development in some diseases,on the other hand,it has protective effect in other cases.Therefore,further studies are required to elucidate the role and mechanism of osteopontin in different diseases.
