摘  要:目的探讨四逆汤复方颗粒剂对小鼠肠道菌群及免疫功能的影响。方法选用100只NIH雌性小鼠,随机分成5组:正常组(生理盐水)、常量组(0.24 g/kg)、超剂量A组(1.2 g/kg)、超剂量B组(2.4g/kg)及超剂量C组(4.8 g/kg),每组20只;每天观察各组小鼠体重、饮食及运动等一般情况;灌胃21 d后,观察各组小鼠的脾指数、胸腺指数、淋巴细胞数、细菌移位现象及肠道菌群等指标。结果四逆汤超剂量B、C组小鼠体重增长缓慢,灌胃14 d后,球杆菌比大于0.333;脾、胸腺指数及淋巴细胞数较正常组明显减少(P<0.05)。常量组及超剂量A组小鼠肠道菌群及免疫相关指标较正常组相比,均无明显差异(P>0.05)。结论 10倍及20倍常量的四逆汤组方颗粒剂可导致小鼠肠道菌群紊乱,免疫功能降低。Objective To investigate effects of Sini prescription granules to the balance of mice intestinal flora. Methods 100 NIH mice were randomly divided into 5 groups, health control(normal saline); normal dose group(0.24 g/kg), overdose group A(1.2 g/kg), overdose group B(2.4 g/kg), overdose group C(4.8 g/kg), each of which contains 20 mice. And then, weights and behaviors of the mice were observed after 21 days gastric lavage, and lymphocyte count, bacterial translocation and intestinal flora of the mice were investigated. ResultsMice with Sini prescription granules in 2.4 g/kg and 4.8 g/kg-dosage groups got slow weight growths during the test;gavage after 21 days, intestinal probiotics of the mice decreased significantly than the normal control group. Ball coli came to more than 1/3, the microflora flora balance was destroyed; ileal mucosa became thinner, villous were significantly atrophied and fell off, decrease of the lymphocyte count was significant(P〈0.05). The 0.24 g/kg-dosage and 1.2 g/kg-dosage groups were similar with the normal group. Conclusion Administration of Sini prescription granules in 2.4 g/kg and 4.8 g/kg-dosage after a period of time, there were inhibitory effects to intestinal probiotics.
