摘 要:目的采用高效液相色谱-质谱(HPLC-MS^n)法,分析延胡索饮片的生物碱类成分。方法采用C_(18)色谱柱,乙腈-0.2%乙酸水溶液梯度洗脱,LTQ-Orbitrap线性离子阱高分辨质谱,正离子检测模式进行扫描,解析多级质谱数据,进行成分定性分析。结果在正离子检测模式下,延胡索饮片中检测出25种成分,鉴定了18种生物碱类成分,其中10种叔胺碱,8种季胺碱成分。结论建立的液相色谱结合高分辨质谱分析延胡素饮片成分的方法快速简便,可用于复杂成分分析,为延胡索饮片质量控制提供了参考依据。Objective A LC-LTQ-Orbitrap MS/MS method was developed for the study of the alkaloids in traditional Chinese medicine Corydalis yanhusuo. Methods Chromatographic separation was achieved on a C_(18) column with the mobile phase of acetonitrile- water(0.2% acetic acid) and step gradient elution. ALTQ-Orbitraptandem mass spetrometerequantification was equipped with electrospray ionization source was used as detetor and operated in the positive ion mode. Qualitative determination of the alkaloids was identified based on the high-resolution mass information, MS/MS fragmentation behaviors. Results 18 compounds were identified or tentatively characterized from 25 alkaloids in Corydalis yanhusuo, including 10 tertiary alkaloids and 8 quaternary ammonium alkaloids. Conclusion The method is rapid and simple in the qualitative analysis of multiple alkaloids in Corydalis yanhusuo using LC-LTQ-Orbitrap MS/MS. This study could be helpful for the quality control and further phytochemical studies of Corydalis yanhusuo decoction pieces.
