摘  要:为建立鸡白色念珠菌的最佳染色方法,探讨了苏木精-伊红(HE)染色、过碘酸雪夫氏(PAS)染色、Grocott六胺银(GMS)染色、CalcofluorWhite(CFW)荧光染色和CFW+HE复染共5种染色方法对鸡嗉囊白色念珠菌感染的染色效果。结果显示,HE染色组织结构清楚,但菌丝和孢子结构不十分清晰;PAS染色和GMS染色菌丝和孢子结构清晰,但步骤较多,且耗时长;CFW荧光染色简单、特异,菌丝、孢子结构清晰,但组织结构不清晰;CFW荧光染色后、再用HE复染能清晰显示菌丝孢子结构及病料组织结构,是较理想的鸡嗉囊白色念珠菌的染色方法。To establish an optimal staining method for chicken Candidaalbicans,this study compared with the effects of different staining methods (Hematoxylin and eosin stain,Grocott-Oomorimethenamine silver stain,Periodic acid-Schiff stain and Calcofluor White fluorescence stain)for C albicans in chicken crop. The results of hematoxylin and eosin stain showed that the structure of crop was clear completely,but the structure of hyphaeand spores was not clear. The results of Grocott-Gomorimethenamine silver stain and periodic acid-schiff stain showed that the structure of hyphae and spores was clear. However,the disadvantages of these two methods were that staining steps were tedious and time-cosuming. Calcofluor White fluorescence stain was simple and specific. It showed that the structure of hyphae and spores was clear,but the structure of crop was not clear. Therefore,this study adopted a new method by which C albicans were stained with the method of Calcofluor White fluorescence stain and then stained with the method of hematoxylin and eosin stain for chicken crops. This combination staining method was an optimal staining method for C. albican chicken crop.
