摘  要:为了建立须癣毛癣菌快速诊断方法,采集疑似患真菌病的家兔皮屑样品96份,制作涂片,分别进行直接镜检、G rocott六胺银染色、过碘酸雪夫氏染色、Calcofluor W hite荧光染色和定量PCR检测。结果显示,4种常规方法的阳性率依次为69.78%、71.19%、72.92%和84.38%,特异性和阳性预测值没有显著差异,而荧光染色检查的敏感性和阴性预测值明显高于其他3常规种方法(P<0.01)。定量PCR的阳性率略高于荧光染色,但没有显著差异。与其他3种常规方法相比,Calcofluor White荧光染色方法相对简单、快捷、敏感、阳性率高,其阳性率与定量PCR相当,但费用低。由此可见,Calcofluor White荧光染色方法是须癣毛癣菌感染可选择的检测方法。To develop quick and sensitive diagnosis methods,scurf speciments were collected from 96 rabbits suspected to be infected by Trichopflyton mentagrophyte. The scurf smears were examined by direct mieroscopy,Grocott's methenamine silver nitrate stain,periodic acid-Schiff stain,calcofluor white stain and quantitative PCR method. The results showed that positive rates of the 4 conventional methods were 69.78%,71.19%,72.92%and 84.38%,respectively. For the 4 conventional methods,there were no differences in specificity and positive predictive value. The calcofluor white stain was the most sensitivity among those methods and was superior to other methods in negative predictive vahe (P〈0.01).The positive rate of quantitative PeR was slightly higher than that of calcofluor white stain,but there was no significant difference. In eonclusion,calcofluor white stain was easy rapid, sensitive and can give significantly higher rates of detection of rrichophyton mentagrophyte infections compared to the other 3 conventional methods. The positive rate of calcofluor white stain was equivalent to that of PeR,but the cost was lower. Therefore,calcofluor white staining should be a single method of choice for the evaluation of Trichophyton mentagrophyte infections.
