SAA与HP在奶牛活体和离体炎性子宫内膜上皮细胞中表达的研究

摘　　要：为研究奶牛子宫内膜炎时急性期蛋白SAA和HP的分子表达规律,采集奶牛子宫组织,通过病理学研究鉴定出不同炎症强度的子宫组织样本;同时体外培养奶牛子宫内膜上皮细胞,以致炎因子细菌脂多糖(LPS)诱导建立了细胞炎症模型。应用RT-qPCR检测子宫组织和细胞炎症模型中SAA和HP基因的表达变化,以Western-blot检测子宫组织中SAA蛋白的表达变化。结果显示,在子宫组织中,SAA基因与蛋白的表达水平均与子宫内膜炎强度呈正相关,然而HP基因表达水平与组织炎症强度无显著相关性。此外,在子宫内膜上皮细胞体外炎症模型中,SAA基因表达水平与LPS剂量呈正相关,而HP基因表达水平呈负相关。结果表明,子宫组织中和体外培养的奶牛子宫内膜上皮细胞中都能表达SAA和HP。作为急性期反应的生物标记,与SAA相比,HP可能对炎症更敏感,其基因表达更早更迅速。To study molecular expression patterns of acute phase proteins serum amyloid A（SAA） and haptoglobin（HP）in dairy cow＇s endometritis, uterine tissues were collected, and different inflamma- tion magnitude were also identified by pathological research. At the same time, endometrial epithelial cells（EECs） were cultured in vitro, and a cell inflammation model was established through inducing by proinflammation factor LPS. Gene expression of SAAand HP was determined by RT-qPCR in both of uterine tissues and cell inflammation model, and protein expression of SAAin uterine tissues was also detected by Western-blot. The results indicated that the levels of genes＇ expression and protein expression of SAAwere positively related to inflammation magnitude in uterine tissue,and gene expression of HP, however, was not significantly related to inflammation magnitude of tissue. Additionally, correlation between gene expression of SAA in cell inflammation model and LPS dosage was still positive, but that of HP was negative. The results showed that genes expression of SAiand HP could take place in both uterine tissue and cultured EECs in vitro. As biomarkers of acute phase response, HP may be more sensitive to inflammation, and its gene expression was also earlier and faster compared to SAA.
