摘 要:目的从形态及功能两个方面观察补肾益精方(BSYJ)对RCS大鼠视网膜变性损伤的保护作用。方法采用先天性视网膜色素变性RCS大鼠作为动物模型,将36只RCS大鼠随机分为蒸馏水组和BSYJ组,每组18只,分别予蒸馏水及BSYJ水煎剂灌胃(8.8 g·kg-1),每日1次,以SD大鼠作为正常对照组常规饲养。分别于干预7 d、14 d和28 d进行指标观察,采用视觉电生理仪检测各组大鼠视网膜电图变化;HE染色观察视网膜各层细胞结构和形态的变化,并计数外核层的细胞核数目。结果视网膜电图结果显示,与正常大鼠比较,RCS大鼠ERG波幅明显降低,在7周龄时波幅几近消失。与蒸馏水组相比,治疗7 d时,BSYJ组明视ERG,暗视ERG以及Ops波幅略有提高,但差异无统计学意义(P>0.05),在治疗14d时,BSYJ组暗视ERG波幅值较蒸馏水组明显提高(P<0.05),治疗28 d时,BSYJ组明视ERG,暗视ERG以及Ops波幅均较蒸馏水组有明显改善(P<0.05,P<0.01)。病理学检查显示,与SD大鼠相比,随着观察时间延长,RCS大鼠视网膜外核层逐渐变薄,外核层细胞数减少。与蒸馏水组比较,BSYJ组在7 d、14 d、28 d观察时间点视网膜形态改善,外核层细胞核数目明显增加(P<0.05,P<0.01)。结论 BSYJ方对RCS大鼠视网膜变性损伤具有一定的保护作用,可能延缓病情的发展过程。OBJECTIVE To investigate the effect of Bushenyijing Decoction(BD) on retinopathy in RCS rats by studying the retinal function and histopathology. METHODS Royal College of Surgeons(RCS) rats were used as an animal model of retinal degeneration. Thirty-six RCS rats were randomly divided into two groups: the distilled water group and the Bushenyijing Decoction(BD) group while SD rats were used as normal control group. The mice of BSYJF group were administrated with Bushenyijing Decoction(BD) solution by gavage(8.8 g·kg-1), and the mice in distilled water group were treated with distilled water by gavage, while the SD mice were fed normally. At the 7th days, 14 thdays and 28 thdays after treatment, their effects were assessed respectively. The flash electroretinogram(FERG) was used to detect the function of the retina. Hematoxylin-eosin(HE) staining was used to evaluate the retinal histopathological changes and the number of photoreceptor nucleiin the outer nuclear layer(ONL). RESULTS The ERG result showed that in contrast with normal control group, the amplitude of RCS rats was significantly de-creased, and it almost disappeared when the RCS rats were 7 weeks old. At 7thday after treatment, in comparison with the distilled water group, the amplitude of photopic ERG, scotopic ERG and Ops of the Bushenyijing Decoction(BD) group was increased slightly, but the difference was not statistically significant(P 〉0.05). At 14 thday after treatment, b-wave amplitudes of scotopic ERG in the BSYJ group was significantly higher than the distilled water group(P〈0.05). At 28 thday after treatment,the amplitudes of scotopic ERG, photopic ERG, and OPs in Bushenyijing Decoction(BD) group were all markedly higher than that in distilled water group(P〈0.05, P〈0.01). It was shown by pathological examination that in comparison with SD mice, the ONL layer in the RCS rats attenuated gradually, and the number of nucleus decreased either.
