摘 要:目的利用基因芯片技术,检测人膀胱尿路上皮癌与正常膀胱组织之间LncRNA表达谱的差异,对差异表达的LncRNA进行分类汇总,分析其生物学信息。方法取人膀胱尿路上皮癌组织与配对的癌旁正常组织3对。分别提取尿路上皮癌组织及正常癌旁组织的总RNA并纯化RNA,与人LncRNA芯片杂交。使用Agilent Genespring GX(Agilent)软件分析尿路上皮癌组织及正常癌旁组织的LncRNA表达情况。结果人膀胱尿路上皮癌组织与配对的癌旁正常组织的LncRNA表达谱差异明显。以P<0.05,上调或下调信号差异大于2倍变化为标准,与正常膀胱组织比较,共筛选出差异表达基因1 122个,其中上调基因734个,下调基因388个。结论膀胱尿路上皮癌的发生发展是一个涉及多基因调控、多条染色体参与的复杂过程。所获得的膀胱癌差异表达LncRNA,尤其差异表达明显的上调基因AK124776、lincRNA-RAB12-1、KRT8P25、RP11-474J18.4、AC000110.1、KRT8P13、KRT8P10、BC072678等、下调基因nc-HOXB9-206、RP11-160A10.2、nc-HOXA11-86、nc-HOXD10-7、nc-HOXB9-205、CES4、nc-HOXD12-3等,考虑上述上调、下调基因对于膀胱癌发生的分子机制及膀胱癌的临床早期诊疗具有重要意义。Objective To analyze the expression profile of LncRNA in normal urinary bladder tissue and bladder urothelial carcinoma,to gather and classify the differently expressed LncRNA,and to analyze the biological information.Methods A total of 3specimens of bladder urothelial carcinomas and3 specimens of normal urinary bladder tissues were collected.Total RNA was isolated,purified and hybridized with human LncRNA.The expressions of LncRNA urothelial carcinomas and adjacent normal tissues were analyzed with Agilent Genespring GX(Agilent)software.Results The expression profile of LncRNA was significantly different between normal urinary bladder tissues and bladder urothelial carcinomas.Compared with normal urinary bladder tissues,the bladder urothelial carcinoma had 1 122 differently expressed genes(P0.05),of which734 were upregulated and 388 were downregulated.Conclusions Bladder urothelial carcinoma involves the regulation of multiple genes and participation of multiple chromosomes.Differently expressed LncRNA involve upregulated genes including AK124776,lincRNA-RAB12-1,KRT8P25,RP11-474J18.4,AC000110.1,KRT8P13,KRT8P10,BC072678 and downregulated genes including nc-HOXB9-206,RP11-160A10.2,nc-HOXA11-86,nc-HOXD10-7,nc-HOXB9-205,CES4,nc-HOXD12-3and so on.Further research into these LncRNA will help to clarify the mechanism of bladder urothelial carcinoma,and to guide the early diagnosis and treatment.
