摘  要:本文通过研究丁香酚(Eugenol)与黄芩(Scutellaria baicalensiz)、葡萄籽(Grape seeds)、五倍子(Chinesegall)、柠檬(Lemon)的提取物对变形链球菌(Streptococcus mutans)及其生物被膜的药效,旨在从中筛选出具有防龋潜能的提取物。采用微量肉汤法测定5种天然药物的最低抑菌浓度(Minimal inhibitory concentration,MIC)、最低杀菌浓度(Minimal bactericidal concentration,MBC)、最低抑制生物被膜浓度(Minimal biofilm inhibitory concentration,MBIC);微量稀释棋盘法测定任两种提取物联合使用时的分级抑菌浓度指数(Fractional inhibitory concentration indexes,FICI);构建生物被膜体外模型,测得提取物对生物被膜生成量的影响;计算各提取物作用生物被膜后被膜内的存活菌数量,并绘制杀菌曲线。丁香酚/柠檬的MBIC/MIC为1~2,在5种提取物中较小;丁香酚、五倍子及柠檬提取物在抑制生物被膜生成方面的效果明显优于黄芩和葡萄籽提取物;杀菌曲线显示柠檬、丁香酚杀菌效果较好,且二者具有一定的协同作用,可以将二者联合使用防龋。To select the extract which owed its anti-caries capacity via pharmacodynamics research of five common plant extracts eugenol,scutellariae baicalensis ,grape seeds, Chinese gall,and/emon extracts agaist Streptococcus mutans and its biofilm,MIC ( minimal inhibitory concentrations), MBC (minimal bactericidal concentration), and MBIC (minimal biofilm inhibitory concentration) were examined by micro-broth dilution method. FIC (fractional inhibitory concentration) indexes in any two extracts were tested by check- erboard method. Biofilm model in vitro was constructed and the effect of extracts were determined by the calculation of biofilm forma- tion quantity. By counting the live bacteria on the agar plates, calculated the survival bacteria number in biofilms after the biofilms were treated by different extracts drawing the sterilization curve. The MBIC/MIC of Eugenol and Lemon are 1- 2, which are lower than those of other extracts. The result exhibited Chinese gall,Eugenol and Lemon extracts had better inhibiton effect on biofilm than the other two extracts. According to the sterilization curve, Lemon and Eugenol are sterilized effectively, in the meantime they had synergy effect,which suggested that they could be combined in using.
