摘 要:目的探讨全氟辛烷磺酸盐(PFOS)对成人神经母瘤细胞(SH-SY5Y)的凋亡效应及可能的机制。方法实验中PFOS剂量设置为200、150、100、50、10、1μmol/L和DMSO(对照组)。体外培养SH-SY5Y细胞,以不同浓度PFOS(1、10、50、100、150、200μmol/L)处理细胞。染毒24 h和48 h后用CCK8法检测SH-SY5Y细胞活性,应用Annexin-V FITC/PI双染法结合流式细胞术检测细胞凋亡,紫外和荧光分光光度法检测SH-SY5Y细胞超氧化物歧化酶(SOD)、谷胱甘肽还原酶(GSH)、总活性氧(ROS)和丙二醛(MDA)的影响。结果 PFOS暴露降低了SH-SY5Y细胞活性,并呈明显的剂量效应关系;PFOS导致SH-SY5Y细胞凋亡;紫外和荧光分光光度法结果显示PFOS引起SH-SY5Y细胞中SOD和GSH水平下降,与对照组相比,最高剂量组PFOS引起GSH含量从8.00±0.12 nmol/mg蛋白降至5.89±0.90 nmol/mg蛋白;同时引起总活性氧(ROS)和丙二醛(MDA)水平上升,与对照组相比,最高剂量组PFOS引起MDA含量从1.92±0.17 nmol/mg蛋白显著上升至6.95±0.26 nmol/mg蛋白。结论PFOS可能通过对SH-SY5Y细胞产生氧化应激损伤从而对SH-SY5Y细胞产生毒性效应。Objective Explore the apoptosis effects of PFOS on SH-SYSY cells and its mechanism. Methods The experiment was divided into seven groups,the control (DMSO) and the PFOS groups (1,10,50,100,150,200 μmol/ L) .The cell viability and time-efficiency of SH-SYSY were determined by CCK8 ,the apoptosis rate were detected by Annex- in-V FITC/PI double-staining,respectively.The effects of PFOS on oxidative stress damage in SH-SYJY cells were detected by UV spectrophotometry and fluorescence methods. Results PFOS treatment significantly decreased SH-SYSY cell via- bility in a dose dependent manner.The apoptosis effects were observed in SH-SYSY cells obviously after PFOS exposure 48 h ; The results of UV and fluorescence spectrophotometry showed that PFOS induced SH-SYSY cell superoxide dismutase (SOD) and glutathione reductase (GSH) decreased,compared with the control,and the highest group of PFOS decreased GSH from 8±0.12 nmol/mg protein to 5.89±0.9 nmol/mg protein.While the total reactive oxygen species (ROS) and malondialdehyde (MDA) levels increased ,compared with the control, the highest group of PFOS elevated MDA from 1.92± 0. 17 nmoL/mg protein to 6.95±0.26 nmol/mg protein. Conclusion PFOS produced oxidative stress damage in SH-SYSY cells, which may be the mechanisms of neurotoxicity induced by PFOS.
